Applicable Applications for anti-MAP2K5 antibody
Western Blot (WB)
Application Notes
Quality Control: Western blots performed on each lot.
WB: 1:1000
Antigen
Phosphopeptide corresponding to amino acid residues surrounding the phospho-Ser311, Thr315 of MEK5.
Immunogen Information
Synthetic phospho-peptide corresponding to amino acid residues surrounding Ser311/Thr315 conjugated to KLH
Immunogen Species
Rat
Reactivity Assumed Based on 100% Sequence Homology
Bovine, canine, chicken, human, mouse, non-human primates, Xenopus and zebra fish
Species Reactivity Note
The antibody has been directly tested for reactivity in Western blots with rat tissue. It is anticipated that the antibody will react with mouse, human, chicken, bovine, canine, non-human primate, Xenopus, and Zebrafish tissues based on the fact that these species have 100% homology with the amino acid sequence used as antigen.
Biological Significance
MEK5 (also known as MKK5) is a dual specificity serine/threonine protein kinase belonging to the MAP kinase kinase family. MEK5 has been shown to specifically activate ERK5 (Zhou et al., 1995) whereas MEK5 itself is regulated by MEKK3 (Chao et al., 1999). An important link between MEK5 and metastatic prostrate cancer has been demonstrated (Mehta et.al., 2003). Dual phosphorylation of Ser311 and Thr315 have been implicated in cell proliferation (Cameron et al., 2004).
Preparation and Storage
Store at -20 degree C; stable for at least one year.

NCBI Description

kinase involved in signal transduction in the cell [RGD, Feb 2006]

Uniprot Description

Function: Acts as a scaffold for the formation of a ternary MAP3K2/MAP3K3-MAP3K5-MAPK7 signaling complex. Activation of this pathway appears to play a critical role in protecting cells from stress-induced apoptosis, neuronal survival and cardiac development and angiogenesis

By similarity. Ref.1

Catalytic activity: ATP + a protein = ADP + a phosphoprotein.

Cofactor: Magnesium.

Subunit structure: Interacts with PARD6A, MAP3K3 and MAPK7. Forms a complex with SQSTM1 and PRKCZ or PRKCI

By similarity.

Subcellular location: Isoform Beta: Cytoplasm › cytosol. Isoform Alpha-1: Membrane. Note: The alternatively spliced exon in alpha isoform resembleconserved sequences that mediate interactions with the cytoskeleton, thereby explaining the differential localization.Isoform Alpha-2: Membrane. Note: The alternatively spliced exon in alpha isoform resembleconserved sequences that mediate interactions with the cytoskeleton, thereby explaining the differential localization.

Tissue specificity: MEK5 beta is ubiquitously distributed with highest levels in the liver, whereas MEK5 alpha is expressed only in liver and brain.

Domain: Binds MAP3K2/MAP3K3 and MAPK7 via non-overlapping residues of the OPR domain. This domain also mediates interactions with SQSTM1 and PARD6A

By similarity.

Post-translational modification: Activated by phosphorylation on Ser/Thr by MAP kinase kinase kinases

By similarity.

Sequence similarities: Belongs to the protein kinase superfamily. STE Ser/Thr protein kinase family. MAP kinase kinase subfamily.Contains 1 OPR domain.Contains 1 protein kinase domain.

Product Notes

The MAP2K5 map2k5 (Catalog #AAA502075) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-Phospho-Ser311, Thr315 MEK5 reacts with Rat and may cross-react with other species as described in the data sheet. AAA Biotech's MEK5 (Ser311, Thr315) can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Quality Control: Western blots performed on each lot. WB: 1:1000. Researchers should empirically determine the suitability of the MAP2K5 map2k5 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MEK5 (Ser311, Thr315), Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
