Biological Activity
Specific activity is > 1000 units/mg, and is defined as the amount of enzyme that cleaves 1umole of oxalacetate and beta-NADH to L-malate and beta-NAD per minute at pH8.0 at 37C.
Assay
1. Prepare a 180ul assay buffer into a suitable container. The concentrations are 100 mM potassium phosphate (pH8.0), 0.5mM beta-nicotinamide adenine dinucleotide, reduced form, 0.5mM oxalacetic acid
2. Equilibrate to 37C and monitor at A340nm until the value is constant using a spectrophotometer.
3. Add 20ul of recombinant MDH protein 0.25ug/ml in assay buffer.
4. Mix by inversion and record the decrease at A340nm for 5 minutes.
Preparation and Storage
Can be stored at 4 degree C short term (1-2 weeks).
For long term storage, aliquot and store at -20 degree C or -70 degree C.
Avoid repeated freezing and thawing cycles.

Malate dehydrogenase (mdh) belongs to the LDH/MDH superfamily and MDH type 1 family. This enzyme catalyzes the conversion of malate into oxaloacetate (using NAD+) and vice versa (this is a reversible reaction). Malate dehydrogenase is also involved in gluconeogenesis, the synthesis of glucose from smaller molecules. Recombinant E Coli mdh protein, fused to His-tag at N-terminus, was expressed in E Coli and purified by using conventional chromatography techniques.
