Activity Data (Figure. Cell proliferation of Raw264.7 cells after stimulated with SCF.Stem cell factor (also known as SCF, KIT-ligand, KL, or steel factor) is a cytokine that binds to the c-KIT receptor (CD117). SCF can exist both as a transmembrane protein and a soluble protein. This cytokine plays an important role in hematopoiesis (formation of blood cells), spermatogenesis, and melanogenesis. SCF has been shown to stimulate the proliferation of Raw264.7 cells. To test this effect, Raw264.7 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach later, replaced with serum-free overnight, then the medium was replaced with 2% serum standard DMEM containing various concentrations of recombinant mouse SCF. After incubated for 120h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of Raw264.7 cells after incubation with SCF for 120h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8) assay after incubation with recombinant SCF for 120h. The result was shown in Figure 2. Activity Data (Platelet Derived Growth Factor Subunit A (PDGFA) is a member of Platelet-derived growth factor (PDGF) family which plays a significant role in blood vessel formation, the growth of blood vessels from already-existing blood vessel tissue, mitogenesis, i.e. proliferation, of mesenchymal cells such as fibroblasts, osteoblasts, tenocytes, vascular smooth muscle cells and mesenchymal stem cells as well as chemotaxis, the directed migration, of mesenchymal cells. Besides, Platelet Derived Growth Factor Receptor Alpha (PDGFRa) has been identified as an interactor of PDGFA, thus a binding ELISA assay was conducted to detect the interaction of recombinant human PDGFA and recombinant human PDGFRa. Briefly, PDGFA were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100mul were then transferred to PDGFRa-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-PDGFA pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of PDGFA and PDGFRa was shown in Figure 1, and this effect was in a dose dependent manner.Figure. Western Blot (WB) (Figure 1. Western blot analysis of GAS 6 using anti-GAS 6 antibody (MBS1750446).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: rat brain tissue lysates,Lane 2: mouse brain tissue lysates.After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-GAS 6 antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for GAS 6 at approximately 75KD. The expected band size for GAS 6 is at 79KD. )
