(Perforin 1 (PRF1) is a pore forming cytolytic protein found in the granules of cytotoxic T lymphocytes (CTLs) and NK cells. Upon degranulation, perforin binds to the target cell's plasma membrane, and oligomerises in a Ca2+ dependent manner to form pores on the target cell. Perforin is thought to act by creating holes in the plasma membrane which triggers an influx of calcium and initiates membrane repair mechanisms. These repair mechanisms bring perforin and granzymes into early endosomes. Besides, Granzyme A (GZMA) has been identified as an interactor of PRF1, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse PRF1 and recombinant mouse GZMA. Briefly, PRF1 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100muL were then transferred to GZMA-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-PRF1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. (Figure. The apoptosis of A549 cells by TNFalpha was inhibited by TNFRSF1B.Tumor necrosis factor receptor superfamily member 1B (TNFRSF1B), also known as tumor necrosis factor receptor 2 (TNFR2) and CD120b, is a membrane receptor that binds tumor necrosis factor-alpha (TNFalpha). This protein and TNF-receptor 1 form a heterocomplex that mediates the recruitment of two anti-apoptotic proteins, c-IAP1 and c-IAP2, which possess E3 ubiquitin ligase activity. TNFRSF1B can inhibit cell apoptosis which induced by TNFalpha. Briefly, A549 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach, replaced with serum-free overnight, then the medium was replaced with 2% serum standard DMEM including 1mug/mL TNFalpha and various concentrations of recombinant human TNFRSF1B. After incubated for 96h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate at 37 degree C for 1-4 hours. Apoptosis of A549 cells had been inhibit after incubation with TNFRSF1B for 96h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8) assay after incubation with recombinant TNFRSF1B for 96h. The result was shown in Figure 2. It was obvious that TNFRSF1B significantly suppress cell apoptosis induced by TNFalpha.(A) A549 cells cultured in DMEM contain 1mug/mL TNFalpha and 0.1ng/mL TNFRSF1B for 96h; (B) A549 cells cultured in DMEM only contain 1mug/mL TNFalpha for 96h.)

(Nucb2 (Nucleobindin-2), a calcium-binding protein, is further cleaved into NES1 (Nesfatin-1). NES1 is an anorexigenic peptide and seems to participate in hypothalamic pathways regulating food intake and energy homeostasis, acting in a leptin-independent manner. GADD45A (Growth arrest and DNA damage-inducible protein GADD45 alpha) has been identified as an interactor of Nucb2. Besides, rat GADD45A shares similarities with mouse NES1 in amino acid sequence with the identity of 98%. Thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse NES1 and recombinant rat GADD45A. Briefly, NES1 were diluted serially in PBS, with 0.01%BSA (pH 7.4). (Leptin is a hormone made by adipose cells that helps to regulate energy balance by inhibiting hunger. Many of leptin's effects are mediated through neuropeptide- containing neurons and neuropeptide receptors in the hypothalamus. Although regulation of fat stores is deemed to be the primary function of leptin, it also plays a role in other physiological processes, as evidenced by its multiple sites of synthesis other than fat cells, and the multiple cell types beside hypothalamic cells that have leptin receptors. Besides, Hexokinase 3, White Cell (HK3) has been identified as an interactor of LEP, thus a binding ELISA assay was conducted to detect the interaction of recombinant human LEP and recombinant human HK3. Briefly, LEP were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to HK3-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-LEPpAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of of LEP and HK3 was shown in Figure 1, and this effect was in a dose dependent manner.)

(Interleukin-6 (IL-6), a pro-inflammatory cytokine and an anti-inflammatory myokine, plays important roles in the acute phase reaction, inflammation, hematopoiesis, bone metabolism, and cancer progression. It has been reported that IL-6 significantly increased the MMP-10 protein lever in the human lung cancer A549 cells through the JAK/STAT signaling pathway. (IL17D (Interleukin-17D) belongs to the IL17 (interleukin 17) family, which includes six members (IL-17, IL-17B through IL-17F). The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for GST3 / GST pi at approximately 23KD.
