(Figure. The binding activity of TXLNa with PDIA3.Taxilin Alpha (TXLNa) also known as interleukin-14 (IL-14) is a cytokine that controls the growth and proliferation of both normal and cancerous B cells. TXLNa induces B-cell proliferation, inhibits antibody secretion, and expands selected B-cell subgroups. This interleukin is produced mainly by T cells and certain malignant B cells. Besides, Protein Disulfide Isomerase A3 (PDIA3) has been identified as an interactor of TXLNa, thus a binding ELISA assay was conducted to detect the interaction of recombinant human TXLNa and recombinant human PDIA3. Briefly, TXLNa were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to PDIA3-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-TXLNa pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of TXLNa and PDIA3 was shown in Figure 1, and this effect was in a dose dependent manner.)

(Interleukin 6 (IL-6) is an interleukin that acts as both a pro-inflammatory cytokine and an anti-inflammatory myokine. Current data suggest that direct application of IL-6 on breast cancer cells inhibits proliferation in ER-positive (estrogen- receptor- positive) cells through the Jak/Stat3 pathway. To test the inhibitory effect of IL-6 on proliferation of ER-positive MCF-7 cell line, cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of IL-6. After incubated for 96h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then measure the absorbance at 450nm using a microplate reader after incubating the plate for 1-4 hours at 37oC.The inhibitory effect of IL-6 on cell proliferation of MCF-7 cells observed by inverted microscope and detected by CCK-8 was shown in Figure 1 and Figure 2 respectively (Dose-dependent effect was not detected in this case).)

(IL17RB (Interleukin-17 receptor B) is a receptor for the proinflammatory cytokines IL17B (Interleukin-17B) and IL17E (Interleukin-17E). This receptor has been shown to mediate the activation of NF-kappaB and the production of IL8 induced by IL17E. Thus a binding ELISA assay was constructed to detect the association of recombinant human IL17RB with recombinant human IL17B. Briefly, IL17RB were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IL17B-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IL17RB pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C.
