(PHA-793887 is a novel pan-cdk inhibitor, including cdk1, cdk2, cdk4, cdk5, cdk7, and cdk9 with IC50 in the 5 to 140 nM range. The sample well of each lane was loaded with 50ug of sample under reducing conditions. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. (TSU-68 (SU6668) has significant antitumor activity against many types of tumor xenograft explants in athymic mice. TSU-68 (SU6668) inhibits angiogenesis through several mechanisms, including the induction of apoptosis in vascular endothelial cells and tumor cells, resulting in perturbing existing tumor vascular function and also inhibiting newly synthesis angiogenesis for the tumor growth. TSU-68 (SU6668) has the effect on blocking the tyrosine kinase phosphorylation of the receptors not only for VEGF but also for PDGF and bFGF, and inhibiting angiogenesis. Studies using in vivo experimental models of cancers such as colon cancer have reported that TSU-68 (SU6668) has the effect on inhibiting tumor proliferation and metastasis through the inhibition of tumor angiogenesis. In vitro TSU-68 (SU6668) does not influence the proliferation of ovarian cancer cells. The administration of TSU-68 (SU6668) does not influence the concentration of VEGF or PDGF in the ascetic fi‚uid of mice with peritoneal dissemination of ovarian cancer. TSU-68 (SU6668) does not act directly on VEGF or PDGF, but binds to their receptors, thereby inhibiting their phosphorylation. [1][2])

Western Blot (WB)
(Western blot analysis of CD68 expression in Jurkat cell lysate (MBS179385).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CD68 monoclonal antibody overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for CD68 )
