Principle of the Assay: Human TRAILR3/TNFRSF10C ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A a monoclonal antibody from mouse specific for TRAILR3/TNFRSF10C has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: A26-A236) and test samples are added to the wells, a biotinylated detection a biotinylated polyclonal antibody from goat specific for TRAILR3/TNFRSF10C is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human TRAILR3/TNFRSF10C amount of sample captured in plate.

Background: TRAILR3/TNFRSF10C is a member of the TNF-receptor superfamily. This receptor contains an extracellular TRAIL-binding domain and a transmembrane domain, but no cytoplasmic death domain. This receptor is not capable of inducing apoptosis, and is thought to function as an antagonistic receptor that protects cells from TRAIL-induced apoptosis. TRAILR3 gene was found to be a p53-regulated DNA damage-inducible gene. The expression of this gene was detected in many normal tissues but not in most cancer cell lines, which may explain the specific sensitivity of cancer cells to the apoptosis-inducing activity of TRAIL.
