IFN-gamma is a dimerized soluble cytokine and the only member of the type II interferon. In humans, IFN-gamma is encoded by the IFNG gene and is critical for innate and adaptive immunity against viral, some bacterial and protozoal infections. IFN-gamma is produced predominantly by natural killer and natural killer T cells as part of the innate immune response, and by CD4 Th1 and CD8 cytotoxic T lymphocytes effector T cells once antigen-specific immunity develops as part of the adaptive immune response. IFN-gamma is also produced by non-cytotoxic innate lymphoid cells.
IFN-gamma interacts with its receptors interferon gamma receptor 1 and interferon gamma receptor 2 and activates them. IFN-gamma binding to the receptors activates the JAK-STAT pathway. IFN-gamma also binds to the glycosaminoglycan heparan sulfate and inhibits its biological activity. IFN-gamma is used to treat Chronic Granulomatous Disease and Osteopetrosis, also has potential as cancer immunotherapy to improve survival in Bladder Carcinoma, Melanoma, and Ovarian Carcinoma.
