(ELISA of anti-CD96 antibody on CD96-Fc fusion protein. Binding curve of the rabbit chimeric version of the anti-CD96 antibody 6A6 (MBS488664; blue line) to an ELISA plate coated with mouse CD96-Fc fusion protein (Pr00230-1.9) at a concentration of 5ug/ml. A 3-fold serial dilution from 10000 to 1.7 ng/ml was performed using MBS488664. For signal detection, a 1:4000 dilution of anti-rabbit IgG1 HRP (BioRad) antibody was used.)

CD96: May be involved in adhesive interactions of activated T and NK cells during the late phase of the immune response. Promotes NK cell-target adhesion by interacting with PVR present on target cells. May function at a time after T and NK cells have penetrated the endothelium using integrins and selectins, when they are actively engaging diseased cells and moving within areas of inflammation. Defects in CD96 are a cause of C syndrome (CSYN); also called Opitz trigonocephaly syndrome. This syndrome is characterized by trigonocephaly and associated anomalies, such as unusual facies, wide alveolar ridges, multiple buccal frenula, limb defects, visceral anomalies, redundant skin, psychomotor retardation and hypotonia. A chromosomal aberration involving CD96 has been found in a patient with C syndrome. Translocation t(3;18)(q13.13;q12.1). CD96 gene was located at the 3q13.13 breakpoint. Precise structural analysis around the breakpoint showed that the gene was disrupted by the translocation in exon 5, probably leading to premature termination or loss of expression of CD96 protein. No gene was detected at the chromosome 18 breakpoint. 2 isoforms of the human protein are produced by alternative splicing.

AAA Biotech's CD96 can be used in a range of immunoassay formats including, but not limited to, Blocking (BL), Flow Cytometry (FC/FACS). This antibody blocks murine CD96-CD155 interaction, and has been shown to bind to the first Ig domain of CD96 and compete with CD155 binding (Roman Aguilera et al, 2018). This antibody displays significant anti-metastatic activity in four experimental lung metastases models and one spontaneous tumour metastasis model (Roman Aguilera et al, 2018), with a potency greater than the anti-CD96 monoclonal antibodies 3.3 and 8B10. The binding specificity of this antibody has been determined in flow cytometric analysis of HEK-293 cells transfected with a range of CD96 Ig domain chimeric constructs (Roman Aguilera et al, 2018). Researchers should empirically determine the suitability of the CD96 cd96 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
