Rabbit anti-Mouse PD-1H Monoclonal Antibody | anti-PD-1H antibody
Anti-PD-1H [MH5A]
Immunofluorescence (IF)
(Immunofluorescence staining of fixed mouse splenocytes with anti-PD-1H antibody MH5A (MBS488594) Immunofluorescence analysis of paraformaldehyde fixed mouse splenocytes permeabilized with 0.15% Triton and stained with the chimeric mouse IgG1 version of MH5A () at 10ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (2ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom , DAPI, merged channels and an isotype control. The isotype control was stained with an anti-unknown specificity antibody followed by Alexa Fluor 488 secondary antibody.)
ELISA (EIA)
(ELISA of anti-VISTA antibody on VISTA-Fc fusion protein. Binding curve of the rabbit chimeric version of the anti-VISTA antibody MH5A (; blue line) to an ELISA plate coated with mouse VISTA-Fc fusion protein (Pr00164-1.9) at a concentration of 5ug/ml. A 3-fold serial dilution from 10000 to 1.7 ng/ml was performed using . For signal detection, a 1:4000 dilution of anti-rabbit IgG1 HRP (BioRad) antibody was used.)
The antibody was generated by immunising Armenian hamsters with mPD-1HIg.
Note on publication: Describes the generation of the antibody and characterisation of its theraputic function in models of graft-versus-host disease.

Product Notes
The PD-1H vsir (Catalog #AAA488595) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-PD-1H [MH5A] reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's PD-1H can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Immunohistochemistry (IHC), Blocking (BL), Therapeutics. The antibody has been labelled with biotin and used to stain formalin-fixed tissues from wild-type C57BL/6 mice at 6 weeks of age. This showed specific staining for PD-1H in the T cell zone and marginal zone. The antibody stains P815 cells transfected with full-length PD-1H plasmid, but not control plasmid. Antibody specificity has been confirmed by ELISA. A single 200-ug dose of MH5A prevented aGVHD in mouse models, caused a reduction in accumulation and expansion of CD8+ and CD4+ T cells in spleens and livers and greatly reduced numbers of inflitrating T cells in all aGVHD target tissues examined. Few mAbs targeting a single cosignalling molecule have such a potent preventative effect (Flies D et al, 2011). Researchers should empirically determine the suitability of the PD-1H vsir for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PD-1H, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
