Western Blot (WB)
(Western Blot using anti-CD53 antibody HD77 (MBS488701) Human lymph node (A), human thymus (B) and human spleen (C) tissue lysates (35ug protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of HD77 (MBS488701) at 0.1ug/ml, 0.3ug/ml and 0.5ug/ml, respectively, before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.)

Flow Cytometry (FC/FACS)
(Flow cytometry using the Anti-CD53 antibody HD77 (MBS488701). Paraformaldehyde fixed Daudi cells were stained with anti-unknown specificity antibody (MBS488149; isotype control, black line) or the rabbit IgG version of HD77 (MBS488701, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.)
This antibody was raised by immunising mice with human leukemia cells.

The protein encoded by this gene is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is a cell surface glycoprotein that is known to complex with integrins. It contributes to the transduction of CD2-generated signals in T cells and natural killer cells and has been suggested to play a role in growth regulation. Familial deficiency of this gene has been linked to an immunodeficiency associated with recurrent infectious diseases caused by bacteria, fungi and viruses. AAA Biotech's CD53 can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), Immunohistochemistry (IHC), Immunofluorescence (IF), Western Blot (WB), Immunoprecipitation (IP). This antibody has applications in the detection of normal and neoplastic white blood cells. This antibody has been used to stain tumour and mononuclear cells for CD53 expression in flow cytometric analysis of samples from ovarian carcinoma patients (Marme et al, 2002), and cells of the colon carcinoma line HT29 (Koretz et al, 1992). In immunohistochemical analysis of frozen tissue sections, this antibody has been shown to react with osteoclasts and osteoclast-like giant cells in malignant fibrous histiocytoma and in benign giant cell tumor of tendon sheath (Mechtersheimer et al, 1990). This antibody has been used to re-immunoprecipitate CD53 from PKCalpha immunoprecipiates (Zhang et al, 2001). Researchers should empirically determine the suitability of the CD53 cd53 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
