Testing Data

(Recombinant DmTet (1634-1986, 2601-2708) activity assay 500 nM of 30 nt ssDNA oligos with a 5mCpG site were incubated with 0 ug, 0.625 ug, 1.25 ug, 2.5 ug or 5 ug DmTet (1634-1986 and 2601-2708), respectively, in a 20 ul reaction system at 34 degree C for 1 hour. Sample was concentrated to 10 ul, and 1 ul was spotted onto nylon membrane. Anti-5hmC Ab and anti-5CaC Ab were used to detect products. DmTet (1634-1986 and 2601-2708) activity assay was detected by Dot-Blot.)

Background: Ten Eleven Translocation (TET) family proteins are cytosine oxygenases that catalyze the conversion of 5-methylcytosine (5mC) into 5-hydroxymethylcytosine (5hmC). 5hmC can be further oxidized into 5-formylcytosine (5fC) and 5-carboxylcytosine (5caC) by TET proteins.
