Dot Blot (DB)

(Dot Blot analysis for recombinant TET3 (824-1795) protein. 250 ng of 30 base oligonucleotide (contains a 5mCpG) was incubated with 0 ug, 312.5 and 625 ng TET3 protein, respectively, in buffer containing 50 mM HEPES pH 7.9, 50 ?M Fe(NH4)2(SO4)2, 1 mM ascorbate, 100 uM alpha-ketoglutarate, 200 uM ATP, 1 mM TCEP at room temperature for 1 hour. Sample was concentrated to 5 ul, then 1 ul sample was spotted onto positively charged nylon membrane. 5-hmC antibody was used to detect the generation of products.)

Short Description: Recombinant TET3 (824-1795) protein that includes amino acids 824-1795 of human TET3 protein (accession number NP_001274420.1) was expressed in Sf9 cells and contains an N-terminal 6xHis-tag and DYKDDDDK-Tag with a molecular weight of 111.3 kDa. The recombinant protein is >30% pure by SDS-PAGE. It is suitable for use in cytosine oxygenase assays, enzyme kinetics, inhibitor screening, and selectivity profiling.

Background: TET (Ten Eleven Translocation) family proteins are cytosine oxygenases that catalyze the conversion of 5-methylcytosine (5mC) into 5-hydroxymethylcytosine (5hmC). 5hmC can be further oxidized into 5-formylcytosine (5fC) and 5-carboxylcytosine (5caC) by TET proteins. Methylation at the C5 position of cytosine is an epigenetic modification of the mammalian genome that plays an important role in transcriptional regulation. TET proteins are responsible for initiating the enzymatic deamination process leading to cytosine demethylation. TET3 plays an important role in DNA methylation reprogramming processes in the mammalian zygote. TET3 may also contribute to somatic cell nuclear reprogramming during animal cloning. Both of CXXC domain and catalytic domain are important for TET3 regulating key genes of Xenopus eye and neural development.

Members of the ten-eleven translocation (TET) gene family, including TET3, play a role in the DNA methylation process (Langemeijer et al., 2009 [PubMed 19923888]).[supplied by OMIM, Nov 2010]

TET3: Dioxygenase that catalyzes the conversion of the modified genomic base 5-methylcytosine (5mC) into 5- hydroxymethylcytosine (5hmC) and plays a key role in epigenetic chromatin reprogramming in the zygote following fertilization. Conversion into 5hmC initiates a process leading to cytosine demethylation through deamination into 5-hydroxymethyluracil (5hmU) and subsequent replacement by unmethylated cytosine by the base excision repair system. In zygotes, DNA demethylation occurs selectively in the paternal pronucleus before the first cell division, while the adjacent maternal pronucleus and certain paternally-imprinted loci are protected from this process. Participates in DNA demethylation in the paternal pronucleus by mediating conversion of 5mC into 5hmC. Does not mediate DNA demethylation of maternal pronucleus because of the presence of DPPA3/PGC7 on maternal chromatin that prevents TET3-binding to chromatin. Belongs to the TET family. 3 isoforms of the human protein are produced by alternative splicing.
