Principle
Mouse SPARCL1 ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for SPARCL1 has been precoated onto 96-well plates. Standards(Expression system for standard: NS0, Immunogen sequence: I17-F650) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for SPARCL1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse SPARCL1 amount of sample captured in plate.
Assay Procedure Step 1
Aliquot 0.1ml per well of the 20000pg/ml, 10000pg/ml, 5000pg/ml, 2500pg/ml, 1250pg/ml, 625pg/ml, 312pg/ml mouse SPARCL1 standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffer into the control well (Zero well). Add 0.1ml of each properly diluted sample of mouse cell culture supernatants, serum or plasma (heparin, EDTA, citrate) to each empty well. See "Sample Dilution Guideline" for details. We recommended that each mouse SPARCL1 standard solution and each sample be measured in duplicate.

Preparation and Storage
Store at 4 degree C for 6 months, at -20 degree C for 12 months. Avoid multiple freeze-thaw cycles. (Shipped with wet ice.)

SPARCL1 (SPARC-Like Protein 1), also known as HEVIN, is a protein that in humans is encoded by the SPARCL1 gene. It is mapped to 5 E5; 5 50. 55 cM. The cells in high endothelial venules (HEVs) in lymphoid tissues have a plump, almost cuboidal, appearance and support high levels of lymphocyte extravasation from blood, possibly due to the presence of desmosome-like junctions rather than tight junctions in the HEVs. In chronic inflammation, the activated endothelium of nonlymphoid tissues acquires an HEV-like morphology and function. SPARCL1 is highly expressed in HEV and is thought to contribute to the induction or maintenance of features of the HEV endothelium that facilitate lymphocyte migration.
