Principle
Mouse BMPER ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A polyclonal antibody from goat specific for BMPER has been precoated onto 96-well plates. Standards(Expression system for standard: NS0, Immunogen sequence: V34-R685) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for BMPER is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB are used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the mouse BMPER amount of sample captured in plate.
Assay Procedure Step 1
Aliquot 0.1ml per well of the 20000pg/ml, 10000pg/ml, 5000pg/ml, 2500pg/ml, 1250pg/ml, 625pg/ml, 312pg/ml mouse BMPER standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffer into the control well (Zero well). Add 0.1ml of each properly diluted sample of mouse cell culture supernatants, serum or plasma (heparin, EDTA, citrate) to each empty well. See "Sample Dilution Guideline" for details. We recommended that each mouse BMPER standard solution and each sample be measured in duplicate.

Preparation and Storage
Store at 4 degree C for 6 months, at -20 degree C for 12 months. Avoid multiple freeze-thaw cycles. (Shipped with wet ice.)

NCBI Description

This gene encodes a secreted protein that contains five Von Willebrand factor type C domains and a Von Willebrand factor type D domain and a trypsin inhibitory-like domain. The encoded protein binds to bone morphogenetic proteins (BMP) and regulates their activity. Mutation of the related gene in humans causes diaphanospondylodysostosis. [provided by RefSeq, Mar 2013]

Uniprot Description

BMPER: Inhibitor of bone morphogenetic protein (BMP) function, it may regulate BMP responsiveness of osteoblasts and chondrocytes. Defects in BMPER are the cause of diaphanospondylodysostosis (DSD). A rare, recessively inherited, perinatal lethal skeletal disorder. The primary skeletal characteristics of the phenotype include a small chest, abnormal vertebral segmentation, and posterior rib gaps containing incompletely differentiated mesenchymal tissue. Consistent craniofacial features include ocular hypertelorism, epicanthal folds, a depressed nasal bridge with a short nose, and low-set ears. The most commonly described extraskeletal finding is nephroblastomatosis with cystic kidneys, but other visceral findings have been described in some cases.
