Activity Data (Matrix Metalloproteinase 13 (MMP13) is a member of the matrix metalloproteinase (MMP) family. MMP13 has been proposed to participate in aggrecan degradation associated with osteoarthritis and cleavage of type II collagen in osteoarthritic cartilage explants and in tumor progression and metastasis. In addition, it can cleave type I, III, IV, IX, X and XIV collagens and fibronectin. MMP13 is likely to play a crucial role in the modulation of extracellular matrix degradation and cell-matrix interactions. Although gelatin zymography is mainly used for the detection of the MMP2 and MMP9, it also can be used for MMP13 dection. Briefly, various concentrations of MMP13 (1000ng, 500ng, 250ng, 125ng) were denatured by SDS loading buffer, electrophoresed through sodium dodecylsulphat- polyacrylamide gel (SDS-PAGE; 10% gels) containing gelatin (1mg/mL) with nonreducing conditions. After renaturation, incubation and CCB-stained, active MMP13 would hydrolyze gelatin nearby, which was indicated by the white bands on the gel. In this experiment we use heat-denatured MMP13 protein as negative control, and blood sample as positive control. Result: Gelatin hydrolysis by recombinant rat MMP13 was shown in figure 1.)

Activity Data (ARSF (Arylsulfatase F) is a member of the sulfatase gene family, and more specifically, the arylsulfatase subfamily. Sulfatases are essential for the correct composition of bone and cartilage matrix. The activity of this protein, unlike that of arylsulfatase E, is not inhibited by warfarin. Besides, EP300 (E1A binding protein p300) has been identified as an interactor of ARSF, thus a binding ELISA assay was conducted to detect the interaction of recombinant human ARSF and recombinant human EP300. Briefly, ARSF were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to EP300-coated microtiter wells and incubated for 2h at 37 degree C. Activity Data (Deoxyribonuclease I (usually called DNase I) is a nonspecific endonuclease that cleaves DNA preferentially at phosphodiester linkages adjacent to a pyrimidine nucleotide, yielding 5'-phosphate-terminated polynucleotides with a free hydroxyl group on position 3', on average producing tetranucleotides. It acts on single-stranded DNA, double-stranded DNA, and chromatin. DNase I can be activated by bivalent metals such as Mg2 and Ca2 . This endonuclease enzyme is common reagents used in biochemical methods requiring diestion of DNA and recovery of RNA, or where DNA is to be removed without affecting structural proteins or enzymes. For example, DNase I is frequently used to remove template DNA following in vitro transcription, and to remove contaminating DNA in total RNA preparations (especially those from transfected cells that may contain plasmid DNA), used for ribonuclease protection assays, cDNA library contraction, and RT-PCR. Activity Data (Mechanism: MMP9 is a zinc-dependent enzymes capable of cleaving components of the extracellular matrix, which belongs to the matrix metalloproteinase (MMP) family. It is a gelatinase A, 92kDa type IV collagenase which can hydrolyze gelatin under certain conditions. Gelatin zymography is mainly used for the detection of the gelatinases, MMP-2 and MMP-9 and It is extremely sensitive because levels of 10pg of MMP-2 can already be detected. Briefly, various concentrations of recombinant human MMP9 (1000ng, 500ng, 100ng, 10ng) were denatured by SDS loading buffer, electrophoresed through sodium dodecylsulphate- polyacrylamide gel (SDS-PAGE; 10% gels) containing gelatin (1mg/mL) with nonreducing conditions.
