Activity Data (CA2 (Carbonic anhydrase 2) is an enzyme that catalyzes reversible hydration of carbon dioxide. It is essential for bone resorption and osteoclast differentiation and contributes to intracellular pH regulation in the duodenal upper villous epithelium during proton-coupled peptide absorption. It is widely accepted that CA2 also catalyzes hydrolysis of p-Nitrophenyl Acetate. Thus, a hydration assay was conducted to test the catalytic activity of CA2 using 4-Nitrophenyl Acetate (4-NPA) as substrate. Briefly, different concentrations of CA2 were incubated with 1mM 4-NPA in reaction buffer. The absorbance at the wavelength of 400nm was read per hour, and the result was shown in figure 1. It is obvious that CA2 catalyzes hydrolysis of p-Nitrophenyl Acetate.)

Activity Data (Suppressor of cytokine signaling 3 (SOCS3) is a member of the STAT-induced STAT inhibitor (SSI), also known as suppressor of cytokine signaling (SOCS), family that negatively regulates cytokine signal transduction. SOCS3 is feedback inhibitors of the Janus kinase (JAK) and signal transducer and activator of transcription (STAT) signaling pathway. Inhibits cytokine signal transduction by binding to tyrosine kinase receptors including gp130, LIF, erythropoietin, insulin, IL12, GCSF and leptin receptors. SOCS3 also can bind to JAK2 kinase inhibits its kinase activity and nhibits insulin signaling in adipose tissue and the liver. Activity Data (NEU (Sialidase-1) is an enzyme that catalyzes the removal of sialic acid (N-acetylneuraminic acid) moities from glycoproteins and glycolipids. Activity Data (Matrix Metalloproteinase 13 (MMP13) is a member of the matrix metalloproteinase (MMP) family. MMP13 has been proposed to participate in aggrecan degradation associated with osteoarthritis and cleavage of type II collagen in osteoarthritic cartilage explants and in tumor progression and metastasis. MMP13 is likely to play a crucial role in the modulation of extracellular matrix degradation and cell-matrix interactions. In addition, it can cleave type I, III, IV, IX, X and XIV collagens and fibronectin. Thus we have chosed casein-zymography to measure the activity of MMP13. Briefly, various concentrations of MMP13 (10ug, 5ug, 1ug, 0.1ug, 0.01ug) were denatured by SDS loading buffer, electrophoresed through sodium dodecylsulphat-polyacrylamide gel (SDS-PAGE; 15% gels) containing casein (1 mg/mL) with nonreducing conditions. After renaturation, incubation and CCB-stained, active MMP13 would hydrolyze casein nearby, which was indicated by the white bands on the gel. In this experiment we use heat-denatured MMP13 protein as negative control, and trypsase (1ug/mL) as positive control.Result : Casein hydrolysis by recombinant human MMP13 was shown in figure 1.)

Activity Data (Arginase (Arg) is an enzyme that catalyzes the degradation of arginine to produce urea and omithine, which is crucial in the urea cycle. Cell proliferation of ECV-304 cells inhibit by VEGFR2.Vascular Endothelial Growth Factor Receptor 2 (VEGFR2) also known as kinase insert domain receptor acts as a cell-surface receptor for VEGFA, VEGFC and VEGFD. VEGFR2 functions as the primary mediator of vascular endothelial growth factor activation in endothelial cells. Regulation of VEGFR-2 expression appears critical in mitogenesis, differentiation, and angiogenesis. To test the effect on inhibit the VEGF-dependent proliferation of endothelium cells, ECV-304 cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well and allowed to attach, replaced with serum-free overnight, then the medium was replaced with 2% serum standard DMEM including 1mug/mL Vascular Endothelial Growth Factor C (VEGFC) and various concentrations of recombinant human VEGFR2. After incubated for 96h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Proliferation of ECV-304 cells after incubation with VEGFR2 for 96h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8) assay after incubation with recombinant VEGFR2 for 96h. The result was shown in Figure 2. It was obvious that VEGFR2 significantly inhibit cell viability of ECV-304.(A) ECV-304 cells cultured in DMEM, stimulated with 10ng/mL VEGFR2 for 96h; (B) Unstimulated ECV-304 cells cultured in DMEM for 96h.)
