Activity Data (Figure. The binding activity of SPC with EIF2aK3.Surfactant associated proteins (SPC), is one of the pulmonary surfactant proteins. It is a membrane protein which manufactures surfactant. The propeptide of pulmonary surfactant C has an N-terminal alpha-helical segment whose suggested function was stabilization of the protein structure, since the latter can irreversibly transform from its native alpha-helical structure to beta-sheet aggregates and form amyloid fibrils. Besides, Eukaryotic Translation Initiation Factor 2 Alpha Kinase 3 (EIF2aK3) has been identified as an interactor of SPC, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse SPC and recombinant mouse EIF2aK3. Briefly, SPC were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to EIF2aK3-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-SPC pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of SPC and EIF2aK3 was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (Anti-Müllerian hormone (AMH), also named Müllerian inhibiting substance (MIS) belongs to a tissue-specific TGF-beta superfamily growth factor. It can be expressed by male sertoli cells and postnatal testis, and ovarian granulosa cells of females postpartum. AMH expression is critical to sex differentiation at a specific time during fetal development, it appears to be tightly regulated by SF1, GATA factors, DAX1 and FSH. AMH signals through a characteristic receptor consisting of a type I and a type II receptor serine/threonine kinase. Especially the type II receptor is unique and specific receptor for AMH. Besides, Mothers Against Decapentaplegic Homolog 9 (Smad9) has been identified as an interactor of AMH, thus a binding ELISA assay was conducted to detect the interaction of recombinant human AMH and recombinant human (Smad9) Briefly, AMH were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to Smad9-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-AMH pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of of AMH and Smad9 was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (Figure 1. Activity DataActivity: Hexosaminidase A Alpha (HEXa) is a lysosomal enzyme. There are three predominant isoenzymes: hexosaminidase A, B and S. Hexosaminidase A and the cofactor GM2 activator protein catalyze the degradation of the GM2 gangliosides and other molecules containing terminal N-acetyl hexosamines. The enzymes are composed of two alpha and/or beta subunits, which are coded by HEXA and HEXB genes, respectively. Even though the alpha and beta subunits of hexosaminidase A can both cleave GalNAc residues, only the alpha subunit which contains a key residue, Arg-424 is able to hydrolyze GM2 gangliosides. Hexosaminidase A (alpha polypeptide) plays a critical role in the brain and spinal cord (central nervous system). Besides, Hexosaminidase B Beta (HEXB) has been identified as an interactor of HEXA, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse HEXA and recombinant mouse HEXB. Briefly, HEXA were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to HEXB-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-HEXA pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50DL stop solution to the wells and read at 450nm immediately. Activity Data (ENDOG (Endonuclease G) is a mitochondrial enzyme that cleaves DNA at double-stranded (DG)n. (DC)n and at single-stranded (DC)n tracts. This protein also has ribonuclease (RNase) and RNase H activities. It has been reported that EndoG forms complexes with some other proteins, including HSP70 (Heat shock 70 kDa protein). Besides, mouse HSPA1A (Heat shock 70 kDa protein 1A) shares similarities with rat HSP1A1 in amino acid sequence with the identity of 98.44%. Thus, a binding ELISA assay was conducted to detect the association of recombinant rat ENDOG with recombinant mouse HSPA1A. Briefly, ENDOG were diluted serially in PBS with 0.01%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to HSP1A1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-ENDOG pAb, then aspirated and washed 3 times. Activity Data (Figure. The binding activity of ACE with ACTb.Angiotensin-converting enzyme ACE, is a central component of the renin-angiotensin system (RAS), which controls blood pressure by regulating the volume of fluids in the body. It converts the hormone angiotensin I to the active vasoconstrictor angiotensin II. Therefore, ACE indirectly increases blood pressure by causing blood vessels to constrict. ACE inhibitors are widely used as pharmaceutical drugs for treatment of cardiovascular diseases. Besides, Actin Beta (ACTb) has been identified as an interactor of ACE, thus a binding ELISA assay was conducted to detect the interaction of recombinant human ACE and recombinant human ACTb. Briefly, ACE were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100muL were then transferred to ACTb-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-ACE pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. Activity Data (Figure. The binding activity of ODC with TK1.Ornithine decarboxylase (ODC) is an enzyme can catalyze the decarboxylation of ornithine (a product of the urea cycle) to form putrescine. The ornithine decarboxylation reaction catalyzed by ornithine decarboxylase is the first and committed step in the synthesis of polyamines, particularly putrescine, spermidine and spermine. Polyamines are important for stabilizing DNA structure, the DNA double strand-break repair pathway and as antioxidants. Therefore, ornithine decarboxylase is an essential enzyme for cell growth, producing the polyamines necessary to stabilize newly synthesized DNA. Lack of ODC causes cell apoptosis in embryonic mice, induced by DNA damage. Besides, Thymidine Kinase 1, Soluble (TK1) has been identified as an interactor of ODC, thus a binding ELISA assay was conducted to detect the interaction of recombinant human ODC and recombinant human TK1. Briefly, ODC were diluted serially in PBS with 0.01% BSA (pH 7.4). Duplicate samples of 100muL were then transferred to TK1-coated microtiter wells and incubated for 2h at 37 degree C.
