Activity Data (TNFRSF14 (Tumor necrosis factor receptor superfamily member 14) belongs to the tumor necrosis factor receptor superfamily. TNFRSF14 functions in signal transduction pathways that activate inflammatory and inhibitory T-cell immune response. It binds herpes simplex virus (HSV) viral envelope glycoprotein D (gD), mediating its entry into cells. A binding ELISA assay was conducted to detect the association of TNFRSF14 with TNFa. Briefly, TNFRSF14 were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL TNFRSF14 were then transferred to TNFa-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-TNFRSF14 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of TNFRSF14 and TNFa was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (Macrophage migration inhibitory factor (MIF), also known as glycosylation- inhibiting factor (GIF), L-dopachrome isomerase, or phenylpyruvate tautomerase is a protein classified as an inflammatory cytokine. MIF is an important regulator of innate immunity. It involved in cell-mediated immunity, immunoregulation, and inflammation. MIF plays a role in the regulation of macrophage function in host defense through the suppression of anti-inflammatory effects of glucocorticoids. This lymphokine and the JAB1 protein form a complex in the cytosol near the peripheral plasma membrane, which may indicate a role in integrin signaling pathways. Besides, Major Histocompatibility Complex Class II Invariant Chain (MHCDG) has been identified as an interactor of MIF, thus a binding ELISA assay was conducted to detect the interaction of recombinant human MIF and recombinant human MHCDG. Briefly, MIF were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to MHCDG- coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-MIF pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. Activity Data (Interleukin 10 (IL-10), also known as human cytokine synthesis inhibitory factor (CSIF), is an anti-inflammatory cytokine. IL-10 was initially reported to suppress cytokine secretion, antigen presentation and CD4 T cell activation. Further investigation has shown that IL-10 predominantly inhibits lipopolysaccharide (LPS) mediated induction of the pro-inflammatory cytokines TNFalpha, IL-1beta, IL-12, and IFNgamma made by cells such as macrophages and Th1 T cells. Therefore, THP1 cells were cultured in 24 well plates at a concentration of 106 cells/ml and activated by LPS (1ug/mL) in the absence or presence of IL-10 (5ng/mL, 10ng/mL) for 24h and production of IL-1beta was determined in the supernatants by cytokine specific ELISA.Activation of THP1 cells with LPS (1ug/mL) resulted in high levels of production of IL-1beta and strong inhibitory effects of IL-10 (5ng/mL, 10ng/mL) were observed on the production of IL-1beta as shown in Table 1.)

The binding activity of IL22 with IL10Rb.Interleukin 22 (IL22) is a member of a group of cytokines called the IL-10 family or IL-10 superfamily, a class of potent mediators of cellular inflammatory responses. It shares use of IL-10R2 in cell signaling with other members of this family, IL-10, IL-26, IL-28A/B and IL-29. IL-22 is produced by activated NK and T cells and initiates innate immune responses against bacterial pathogens especially in epithelial cells such as respiratory and gut epithelial cells.
