(Growth Regulated Oncogene Beta (GROb) is a small cytokine belonging to the CXC chemokine family which produced by activated monocytes and neutrophils and expressed at sites of inflammation. Hematoregulatory chemokine, which, in vitro, suppresses hematopoietic progenitor cell proliferation. Besides, Dipeptidyl Peptidase IV (DPP4) has been identified as an interactor of GROb, thus a binding ELISA assay was conducted to detect the interaction of recombinant rat GROb and recombinant rat DPP4. Briefly, GROb were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100ul were then transferred to DPP4-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-GROb pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of of GROb and DPP4 was shown in Figure 1, and this effect was in a dose dependent manner.)

Western blot analysis of Lactoferrin using anti-Lactoferrin antibody (MBS1750451).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: human placenta tissue lysates,Lane 2: rat spleen tissue lysates,Lane 3: mouse spleen tissue lysates.After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Lactoferrin antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for Lactoferrin at approximately 85KD. The expected band size for Lactoferrin is at 78KD. )

(Anti-Ly6G staining of murine neutrophils. Ex vivo murine (C57BL6/J) neutrophils from zymosan-elicited peritoneal exudate were stained with anti-Ly6G antibody and analysed by flow-cytometry. Forward-and side-scatter was used to gate on neutrophil population (data not shown) and fluorescence intensity analysed. Left panel shows the signal obtained from cells pre-treated with Fc-blocking reagent and subsequently stained with anti-Ly6G (hybridoma-derived antibody obtained from competitor, clone 1A8,) directly conjugated to PE. Comparison to unstained cells shows that that cells analysed are predominantly Ly6G-positive. Right panel shows staining first with anti-Ly6G or isotype control followed by staining with fluorescently conjugated secondary antibody (goat anti-rat). As a further control, cells were also only stained with secondary antibody. Comparison of the staining pattern obtained using agreeing with findings obtained using hybridoma-derived antibody.)

The binding activity of GM-CSF with CSF2Ra.Granulocyte-macrophage colony-stimulating factor (GM-CSF), also known as colony stimulating factor 2 (CSF2), is a monomeric glycoprotein secreted by macrophages, T cells, mast cells, natural killer cells, endothelial cells and fibroblasts that functions as a cytokine. GM-CSF stimulates stem cells to produce granulocytes (neutrophils, eosinophils, and basophils) and monocytes. It also has some effects on mature cells of the immune system. These include, for example, inhibiting neutrophil migration and causing an alteration of the receptors expressed on the cells surface. GM-CSF signals via signal transducer and activator of transcription, STAT5. In macrophages, it has also been shown to signal via STAT3. Besides, Colony Stimulating Factor 2 Receptor Alpha (CSF2Ra) has been identified as an interactor of GM-CSF, thus a binding ELISA assay was conducted to detect the interaction of recombinant rat GM-CSF and recombinant rat CSF2Ra. Briefly, GM-CSF were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to CSF2Ra-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-GM-CSF pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of GM-CSF and CSF2Ra was shown in Figure 1, and this effect was in a dose dependent manner.)
