The binding activity of MIF with MHCDG.Macrophage migration inhibitory factor (MIF), also known as glycosylation-inhibiting factor (GIF), L-dopachrome isomerase, or phenylpyruvate tautomerase is a protein classified as an inflammatory cytokine. MIF is an important regulator of innate immunity. It involved in cell-mediated immunity, immunoregulation, and inflammation. MIF plays a role in the regulation of macrophage function in host defense through the suppression of anti-inflammatory effects of glucocorticoids. This lymphokine and the JAB1 protein form a complex in the cytosol near the peripheral plasma membrane, which may indicate a role in integrin signaling pathways. Besides, Major Histocompatibility Complex Class II Invariant Chain (MHCDG) has been identified as an interactor of MIF, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse MIF and recombinant mouse MHCDG. Briefly, MIF were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to MHCDG-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-MIF pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of MIF and MHCDG was shown in Figure 1, and this effect was in a dose dependent manner.)

(Vascular endothelial growth factor A (VEGF-A), a glycosylated mitogen, is known to be a vascular permeability factor and an endothelial cell growth factor secreted by the smooth muscle and endothelial cells. It has been reported that VEGF-A induces vascular permeability and growth, promotes monocyte/macrophage migration, and inhibits cell apoptosis and so on. To test the effect of VEGF-A on cell proliferation of ECV304 endothelium cell line, cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of VEGFA. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then measure the absorbance at 450nm using a microplate reader after incubating the plate for 1-4 hours at 37oC .Cell proliferation of ECV304 cells after incubation with VEGFA for 72h observed by inverted microscope was shown in Figure 1.)

(PTHrP (Parathyroid Hormone Related Protein ) is a neuroendocrine peptide which is a critical regulator of cellular and organ growth, development, migration, differentiation and survival and of epithelial calcium ion transport. Regulates endochondral bone development and epithelial-mesenchymal interactions during the formation of the mammary glands and teeth. A binding ELISA assay was conducted to detect the association of PTHrP with PTHR1. Briefly, PTHrP were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL PTHrP were then transferred to PTHR1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-PTHrP pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of PTHrP and PTHR1 was shown in Figure 1, and this effect was in a dose dependent manner.)

The chemotactic effect of NAP3 on THP-1 cells.Neutrophil-activating protein 3 (NAP3) also known as chemokine (C-X-C motif) ligand 1 (CXCL1) is a small cytokine belonging to the CXC chemokine family. NAP3 is expressed by macrophages, neutrophils and epithelial cells,and has neutrophil chemoattractant activity. Thus, chemotaxis assay used 24-well microchemotaxis system was undertaken to detect the chemotactic effect of NAP3 on the human monocytic cell line THP-1. Briefly, THP-1 cells were seeded into the upper chambers (100uL cell suspension, 106 cells/mL in RPMI 1640 with FBS free) and SLC (31.25ng/mL, 62.5ng/mL,125ng/mL and 250ng/mL diluted separately in serum free RPMI 1640) was added in lower chamber with a polycarbonate filter (8um pore size) used to separate the two compartments. After incubation at 37 degree C with 5% CO2 for 1h, the filter was removed, then cells in low chamber were observed by inverted microscope at low magnification (×100) and the number of migrated cells were counted at high magnification (×400) randomly (five fields for each filter). Result shows NAP3 is able to induce migration of THP-1 cells. The migrated THP-1 cells in low chamber at low magnification (×100) were shown in Figure 1. Five fields of each chamber were randomly chosen, and the migrated cells were counted at high magnification (×400). Statistical results were shown in Figure 2. (CCL13 (C-C motif chemokine 13) is a chemotactic factor that attracts monocytes, lymphocytes, basophils and eosinophils, which belongs to the CC chemokine subfamily. It has been reported that CCL13 can induce chemotactic migration of THP-1 cells. Therefore, chemotaxis assay used 24-well microchemotaxis system was undertaken to detect the chemotactic effect of CCL13 on the human monocytic cell line THP-1.
