(Interleukin 15 (IL15) is a widely expressed cytokine that is structurally and functionally related to IL2, which plays an important role in many immunological diseases. IL15 also regulates T and natural killer (NK) cell activation and proliferation. To test the effect of IL15 on cells proliferation of human T lymphocyte cells, Jurkat cells were seeded into triplicate wells of 96-well plates at a density of 10, 000 cells/well in RPMI-1640 with the addition of various concentrations of IL15. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Cell proliferation of Jurkat cells after incubation with IL15 for 72h observed by inverted microscope was shown in Figure 1. The dose-effect curve of IL15 was shown in Figure 2. It was obvious that IL15 significantly promoted cell proliferation of Jurkat cells. The ED50 for this effect is typically 0.7240 to 5.206ng/mL.)

(IL17RD (interleukin-17 receptor D) is a membrane protein belonging to the IL17R protein family, acting as a component of the interleukin-17 receptor signaling complex. Knowing that the interaction between this protein and IL-17R does not require the interleukin, we have conducted a binding a binding ELISA assay to detect the interaction of recombinant human IL17RD with both recombinant human IL17RA and IL17. Briefly, IL17RD were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IL17RA-coated or IL17-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IL17RA pAb and anti-IL17 pAb separately, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution , wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of IL17RD with IL17RA and IL17 was shown in Figure 1 and Figure 2 respectively and this effect was in a dose dependent manner.)
