(IL36G (Interleukin 36 gamma), also known as IL-1F9 and IL-1H1 is a member of the IL-1 family. The receptor for IL36 gamma is reported to be a combination of IL-1 R6/IL-1 R rp2 and IL-1 R3/IL-1 R AcP. Thus we have conducted a binding ELISA assay to detect the interaction of recombinant human IL36G with recombinant human IL1R1. Briefly, IL36G were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IL1R1-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IL1R1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of IL36G with IL1R1 was shown in Figure 1 and this effect was in a dose dependent manner.)

(IL24 (interleukin 24) is a cytokine that belongs to IL10 family. This protein can induce apoptosis selectively in various cancer cells, including ECV304. Thus, inhibition of cell proliferation assay of IL24 was conducted using ECV-304 cells. Briefly, ECV-304 cells were seeded into triplicate wells of 96-well plates at a density of 2,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard 1640 prior to the addition of various concentrations of IL24. After incubated for 48h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Inhibition of ECV-304 cells proliferation after incubation with IIL24 for 48h observed by inverted microscope was shown in Figure 1. Cell viability was assessed by CCK-8 (Cell Counting Kit-8 ) assay after incubation with various concentrations of IL24 for 48h. The mean OD value of ECV-304 assessed by CCK-8 was shown in Figure 2. It was obvious that IL24 significantly decreased cell viability of ECV-304 cells.)

(IL18 (interleukin 18) is a cytokine that shares similarities in structure with IL1, and is produced by macrophages and other cells. IL18 has been reported to stimulate T cells proliferation and NK cell activity, besides, murine IL18 has been found to enchance the production of INFgamma in spleen cells in vitro. Thus, a stimulation assay was conducted to detect the activity of IL18 using murine spleen cells. Briefly, murine spleen cells were seeded into wells of 6-well plates at a density of 2×106 cells/mL in RPMI-1640 with the addition of various concentrations of IL18. After incubation for 48 hours, the concentration of INFgamma in the cell supernatant was detected using an ELISA kit. INFgamma levels in the cell supernatant of spleen cells increased significantly after stimulated with IL18, the data was shown in Figure 1.)

(Interleukin 6 (IL-6) is an interleukin that acts as both a pro-inflammatory cytokine and an anti-inflammatory myokine. Current data suggest that direct application of IL-6 on breast cancer cells inhibits proliferation in ER-positive (estrogen- receptor- positive) cells through the Jak/Stat3 pathway. To test the inhibitory effect of IL-6 on proliferation of ER-positive MCF-7 cell line, cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of IL-6. The binding activity of IL21 with IL2Rg.Interleukin-21 (IL21) is a cytokine that has potent regulatory effects on cells of the immune system, including natural killer (NK) cells and cytotoxic T cells that can destroy virally infected or cancerous cells. This cytokine induces cell division/proliferation in its target cells. IL21 may be a critical factor in the control of persistent viral infections. Besides, Interleukin 2 Receptor Gamma (IL2Rg) has been identified as an interactor of IL21, thus a binding ELISA assay was conducted to detect the interaction of recombinant human IL21 and recombinant human IL2Rg. Briefly, IL21 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IL2Rg-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-IL21 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of IL21 and IL2Rg was shown in Figure 1, and this effect was in a dose dependent manner.)
