Principle of the Assay: The LumiAb Human MCP-1 Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human MCP-1 concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human MCP-1 while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human MCP-1 or Monocyte Chemoattractant Protein-1, also known as C-C Motif Chemokine 2, is a 99 amino acid chemokine protein encoded by the CCL2 gene located at locus 17q11.2-17q12 on chromosome 17. MCP-1 is mainly a chemotactic factor that attracts monocytes and basophils but not neutrophils or eosinophils. Typically, the chemokine is believed to augment monocyte anti-tumor activity and has also been implicated in the pathogenesis of disease characterized by monocytic infiltrates, like psoriasis, rheumatoid arthritis or atherosclerosis. Moreover, MCP-1 may also be involved in the recruitment of monocytes into the arterial wall during the disease process of atherosclerosis. Able to exist as monomeric or homodimeric in equilibrium, MCP-1 can bind to CCR2 and CCR4, as well as being tethered on endothelial cells by glycosaminoglycan (GAG) side chains of proteoglycans. A common post-translational modification that takes place with MCP-1 is processing at the N-terminus which can regulate receptor and target cell selectivity. Deletion of the N-terminal residue converts it from an activator of basophils to an eosinophil chemoattractant.

NCBI Description

This gene encodes a preproprotein that is proteolytically processed to generate two protein products, megakaryocyte potentiating factor and mesothelin. Megakaryocyte potentiating factor functions as a cytokine that can stimulate colony formation of bone marrow megakaryocytes. Mesothelin is a glycosylphosphatidylinositol-anchored cell-surface protein that may function as a cell adhesion protein. This protein is overexpressed in epithelial mesotheliomas, ovarian cancers and in specific squamous cell carcinomas. Alternative splicing results in multiple transcript variants, at least one of which encodes an isoform that is proteolytically processed. [provided by RefSeq, Feb 2016]
