Flow Cytometry (FC/FACS) (Flow-cytometry using the Anti-IL-17F antibody 8F5.1A9 (MBS488659). Mouse splenocytes were stimulated with PMA and Ionomycin in presence of Monensin for 4 hours. The cells were fixed using 2% PFA, permeabilized using 0.5% Triton and stained with anti-Fluorescein IgG antibody (4-4-20; isotype control, black line) or the rabbit IgG version of 8F5.1A9 (MBS488659, blue line) at a dilution of 1:100 for 1h at RT. After washing, bound antibody was detected using a goat anti-mouse IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells were analyzed using a FACSCanto flow-cytometer.)

Uniprot Description

IL17F: Stimulates the production of other cytokines such as IL- 6, IL-8 and granulocyte colony-stimulating factor, and can regulate cartilage matrix turnover. Stimulates PBMC and T-cell proliferation. Inhibits angiogenesis. Defects in IL17F are the cause of familial candidiasis type 6 (CANDF6). CANDF6 is a rare disorder with altered immune responses and impaired clearance of fungal infections, selective against Candida. It is characterized by persistent and/or recurrent infections of the skin, nails and mucous membranes caused by organisms of the genus Candida, mainly Candida albicans. Belongs to the IL-17 family.
