HMT Assay Conditions: 2ug Recombinant Nucleosomes were incubated with different concentrations of SUV420H1 (2- 387) protein in 30ul reaction system containing 50mM Tris-HCl, pH 8.6, 0.02% Triton X-100, 2mM MgCl2, 1mM TCEP, and 50uM SAM for 3 hours at room temperature. Western Blot was used to detect the generation of reaction products.

(Recombinant SUV420H1 (2-387) activity assay 2 ug Recombinant Nucleosomes (H3.1) were incubated with 0 ug (-), 0.2 ug (+), 0.4 ug (++), 0.8 ug (+++) SUV420H1 (2-387) protein, respectively, in 30 ul reaction system containing 50 mM Tris-HCl, pH 8.6, 0.02% Triton X-100, 2 mM MgCl2, 1 mM TCEP, 50 uM SAM for 3 hr at room temperature. 6 ul reaction products were loaded and run on a 12.5% SDS-PAGE gel. Western Blot was used to detect the generation of reaction products (H4K20me3 antibody). SUV420H1 (2-387) only was used as negative control. The Western blot result shows that nucleosomes are much better than histone octamers as the substrate of SUV420H1 (2-387).)
