Related Product Information for EHMT2 recombinant protein
Short Description: The peptide corresponding to amino acids 913-1193 of the G9a protein (accession number NP_006700.3) was expressed in E Coli. The peptide contains the SET domain of G9a that is responsible for methyltransferase activity. Recombinant G9a-SET contains N-terminal GST-Tag with an observed molecular weight of 58.7 kDa. G9a, also known as Euchromatic Histone-lysine N-methyltransferase 2 (EHMT2), is a histone methyltransferase that specifically mono- and dimethylates Lys9 of histone H3 (H3K9me1 and H3K9me2, respectively) in euchromatin. H3K9me represents a specific tag for epigenetic transcriptional repression by initiating recruitment of HP1 proteins to methylated histones. G9a also mediates monomethylation of Lys56 of histone H3 (H3K56me1) in G1 phase and regulates DNA replication by promoting the interaction between histone H3 and PCNA. G9a also weakly methylates Lys27 of histone H3 (H3K27me). In addition, G9a is required for DNA methylation. However, the histone methyltransferase activity is not required for DNA methylation, suggesting that these 2 activities function independently. G9a is most likely targeted to histone H3 by different DNA-binding proteins like E2F6, MGA, MAX and/or DP1. G9a may also methylate histone H1. In addition to histone methyltransferase activity, G9a also methylates non-histone proteins. It has been shown to mediate dimethylation of Lys373 of p53 and also methylates CDYL, WIZ, ACIN1, DNMT1, HDAC1, ERCC6, KLF12 and itself. Recombinant G9a-SET can be used for enzyme kinetics and inhibitor screening studies.

NCBI Description

This gene is located in a gene cluster within the human major histocompatibility complex class III region on chromosome 6. This gene was thought to be two different genes, NG36 and G9a, adjacent to each other but recent publications show that there is only a single gene at this locus. The encoded protein is a histone methyltransferase, which may be involved in repression of transcription. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jan 2014]

Uniprot Description

EHMT2 iso4: Histone methyltransferase. Preferentially methylates 'Lys-9' of histone H3 and 'Lys-27' of histone H3 (in vitro). H3 'Lys-9' methylation represents a specific tag for epigenetic transcriptional repression by recruiting HP1 proteins to methylated histones. Probably targeted to histone H3 by different DNA-binding proteins like E2F6, MGA, MAX and/or DP1. Also methylates histone H1. Belongs to the histone-lysine methyltransferase family. Suvar3-9 subfamily. Part of the E2F6.com-1 complex in G0 phase composed of E2F6, MGA, MAX, TFDP1, CBX3, BAT8, EUHMTASE1, RING1, RNF2, MBLR, L3MBTL2 and YAF2. Interacts with GFI1B, WIZ and EHMT1. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes

The EHMT2 ehmt2 (Catalog #AAA388142) is a Recombinant Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "EHMT2, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
