The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: human placenta tissue lysates,Lane 2: rat spleen tissue lysates,Lane 3: mouse spleen tissue lysates.After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-HBD antigen affinity purified polyclonal antibody at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for HBD at approximately 16KD. The expected band size for HBD is at 16KD. )

(Figure 1. The binding activity of CKB with CKM.Creatine Kinase, Brain (CKB) also known as CK-BB is a creatine kinase. CKB, consists of a homodimer of two identical brain-type CK-B subunits, is a cytoplasmic enzyme involved in cellular energy homeostasis, with certain fractions of the enzyme being bound to cell membranes, ATPases, and a variety of ATP-requiring enzymes in the cell. Besides, Creatine Kinase, Muscle (CKM) has been identified as an interactor of CKB, thus a binding ELISA assay was conducted to detect the interaction of recombinant human CKB and recombinant human CKM. Briefly, CKB were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to CKM-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-CKB pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of CKB and CKM was shown in Figure 1, and this effect was in a dose dependent manner.)

Gelatin zymography is mainly used for the detection of the gelatinases, MMP-2 and MMP-9 and It is extremely sensitive because levels of 10pg of MMP-2 can already be detected. Briefly, various concentrations of recombinant human MMP9 (1000ng, 500ng, 100ng, 10ng) were denatured by SDS loading buffer, electrophoresed through sodium dodecylsulphate- polyacrylamide gel (SDS-PAGE; 10% gels) containing gelatin (1mg/mL) with nonreducing conditions. After renaturation, incubation and CCB-stained, active MMP2 would hydrolyze gelatin nearby, which was indicated by the white binds on the gel. In this experiment we use heat-denatured MMP9 protein as negative control, and blood sample as positive control. Result: Gelatin hydrolysis by recombinant human MMP9 (10-70kd) was shown in figure 1.)

(Figure 1. The binding activity of CYP3A4 with HSPA8.Cytochrome P450 3A4 (CYP3A4) is an important enzyme in the body, mainly found in the liver and in the intestine. It oxidizes small foreign organic molecules (xenobiotics), such as toxins or drugs, so that they can be removed from the body. CYP3A4 is a member of the cytochrome P450 family of oxidizing enzymes and the CYP3A4 is the most common and the most versatile one in drug metabolism. Besides, Heat Shock 70kDa Protein 8 (HSPA8) has been identified as an interactor of CYP3A4, thus a binding ELISA assay was conducted to detect the interaction of recombinant human CYP3A4 and recombinant human HSPA8. Briefly, CYP3A4 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to HSPA8-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-CYP3A4 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C.
