Activity Data (Golgi Protein 73 (GP73) also known as Golgi phosphoprotein 2 or Golgi membrane protein GP73 is a protein that encoded by the GOLM1 gene. The Golgi complex plays a key role in the sorting and modification of proteins exported from the endoplasmic reticulum. It has been observed to be upregulated in response to viral infection. Besides, Dymeclin (DYM) has been identified as an interactor of GP73, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse GP73 and recombinant human DYM. Briefly, GP73 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to DYM-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-GP73 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of of GP73 and DYM was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (IL7 (Interleukin 7) is a hematopoietic growth factor secreted by stromal cells in the bone marrow and thymus. The interaction between IL17 and the IL7 receptor triggers a cascade of signals important for T-cell development within the thymus and survival within the periphery. It is reported that IL-7 acts on both resting and activated T cells, including Jurkat cells. Thus, a proliferation assay of recombinant human IL7 was conducted using Jurkat cells. Briefly, Jurkat cells were seeded into triplicate wells of 96-well plates at a density of 10, 000 cells/well in RPMI-1640 with the addition of various concentrations of IL7. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C. Cell proliferation of Jurkat cells after incubation with IL7 for 72h observed by inverted microscope was shown in Figure 1. The CCK-8 data was shown in Figure 2. It was obvious that IL7 significantly promoted cell proliferation of Jurkat cells.)

Activity Data (Macrophage Colony Stimulating Factor (M-CSF), also known as CSF-1,is a secreted cytokine which influences hematopoietic stem cells to differentiate into macrophages or other related cell types. M-CSF (or CSF-1) is a hematopoietic growth factor that is involved in the proliferation, differentiation, and survival of monocytes, macrophages, and bone marrow progenitor cells. Activity Data (Protein S100B is a member of the S100 family. S100 proteins are EF-hand calcium-binding proteins and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. The chemotactic effect of NAP3 on THP-1 cells.Neutrophil-activating protein 3 (NAP3) also known as chemokine (C-X-C motif) ligand 1 (CXCL1) is a small cytokine belonging to the CXC chemokine family. NAP3 is expressed by macrophages, neutrophils and epithelial cells,and has neutrophil chemoattractant activity. Thus, chemotaxis assay used 24-well microchemotaxis system was undertaken to detect the chemotactic effect of NAP3 on the human monocytic cell line THP-1. Briefly, THP-1 cells were seeded into the upper chambers (100uL cell suspension, 106 cells/mL in RPMI 1640 with FBS free) and SLC (31.25ng/mL, 62.5ng/mL,125ng/mL and 250ng/mL diluted separately in serum free RPMI 1640) was added in lower chamber with a polycarbonate filter (8um pore size) used to separate the two compartments. After incubation at 37 degree C with 5% CO2 for 1h, the filter was removed, then cells in low chamber were observed by inverted microscope at low magnification (×100) and the number of migrated cells were counted at high magnification (×400) randomly (five fields for each filter). Other properties attributed to the cytokine include: the growth promotion and cell differentiation of different types of target cells, influence on bone metabolism, cachexia, neural development, embryogenesis and inflammation. p53 regulated LIF has been shown to facilitate implantation in the mouse model and possibly in humans. To test the effect of LIF on cell proliferation, TF-1 cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well with 1% serum standard 1640 including various concentrations of recombinant human LIF. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then the absorbance at 450nm was measured using a microplate reader after incubating the plate for 1-4 hours at 37 degree C.
