Activity Data (JAK2 (Tyrosine-protein kinase JAK2) is a tyrosine kinase involved in various processes such as cell growth, development, differentiation or histone modifications. JAK2 is considered to associate with some type I receptors, including EPOR (Erythropoietin receptor), therefore participates in cellular signal transduction. Thus a binding ELISA assay was conducted to detect the interaction of recombinant human JAK2 and recombinant human EPOR. Briefly, JAK2 were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL JAK2 were then transferred to EPOR-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-JAK2 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of JAK2 and EPOR was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (Macrophage Colony Stimulating Factor (M-CSF), also known as CSF-1,is a secreted cytokine which influences hematopoietic stem cells to differentiate into macrophages or other related cell types. M-CSF (or CSF-1) is a hematopoietic growth factor that is involved in the proliferation, differentiation, and survival of monocytes, macrophages, and bone marrow progenitor cells. It can also affects macrophages and monocytes in several ways, including stimulating increased phagocytic and chemotactic activity, and increased tumour cell cytotoxicity. The role of M-CSF is not only restricted to the monocyte/macrophage cell lineage. By interacting with its membrane receptor (CSF1R or M-CSF-R encoded by the c-fms proto-oncogene), M-CSF also modulates the proliferation of earlier hematopoietic progenitors and influence numerous physiological processes involved in immunology, metabolism, fertility and pregnancy. Besides, Colony Stimulating Factor Receptor, Macrophage (M-CFS-R) has been identified as an interactor of M-CSF, thus a binding ELISA assay was conducted to detect the interaction of recombinant mouse M-CSF and recombinant mouse MCFSR Briefly, M-CSF were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to M-CFS-R-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-M-CSF pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of of M-CSF and M-CFS-R was shown in Figure 1, and this effect was in a dose dependent manner.)

Activity Data (Protein S100B is a member of the S100 family. S100 proteins are EF-hand calcium-binding proteins and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. Experimental results suggest that the receptor for advanced glycation end products (RAGE) plays important roles in mediating S100 protein-induced cellular signaling. Besides, rat RAGE shares similarities with human RAGE in amino acids sequence with the identity of 80.0%. Thus a binding ELISA assay was conducted to detect the interaction of recombinant rat S100B and recombinant human RAGE. Briefly, S100B were diluted serially in PBS, with 0.01%BSA (pH 7.4). Duplicate samples of 100uL were then transferred to RAGE-coated microtiter wells and incubated for 2h at 37 degree C. Activity Data (Regenerating Islet Derived Protein 3 Beta (REG3b) also known as PAP-I and HIP is bactericidal C-type lectin which acts against several intestinal Gram-positive bacteria and Gram-negative bacteria. The Reg family proteins have been implicated in a range of physiological processes including acting as acute phase reactants, lectins, survival/growth factors for insulin-producing pancreatic beta-cells, neural cells, and epithelial cells of the digestive system. To test the effect of REG3b on cell proliferation of SK-N-SH, SK-N-SH cells were seeded into triplicate wells of 96-well plates at a density of 5,000 cells/well and allowed to attach overnight, then the medium was replaced with serum-free standard DMEM prior to the addition of various concentrations of REG3b. After incubated for 72h, cells were observed by inverted microscope and cell proliferation was measured by Cell Counting Kit-8 (CCK-8). Briefly, 10uL of CCK-8 solution was added to each well of the plate, then measure the absorbance at 450nm using a microplate reader after incubating the plate for 1-4 hours at 37 degree C .Cell proliferation of SK-N-SH cells after incubation with REG3b for 72h observed by inverted microscope was shown in Figure 1.)

Activity Data (Leptin is a hormone made by adipose cells that helps to regulate energy balance by inhibiting hunger. Many of leptin's effects are mediated through neuropeptide- containing neurons and neuropeptide receptors in the hypothalamus. Although regulation of fat stores is deemed to be the primary function of leptin, it also plays a role in other physiological processes, as evidenced by its multiple sites of synthesis other than fat cells, and the multiple cell types beside hypothalamic cells that have leptin receptors. Besides, Hexokinase 3, White Cell (HK3) has been identified as an interactor of LEP, thus a binding ELISA assay was conducted to detect the interaction of recombinant human LEP and recombinant human HK3. Activity Data (Caspase 1 (CASP1) cleaves IL-1 beta between an Asp and an Ala, releasing the mature cytokine which is involved in a variety of inflammatory processes. It also has lots of other function, such as defenses against pathogens, cleaves and activates sterol regulatory element binding proteins (SREBPs), promotes apoptosis. Besides, Interleukin 1 Alpha (IL1a) has been identified as an interactor of CASP1, thus a binding ELISA assay was conducted to detect the interaction of recombinant rat CASP1 and recombinant rat IL1a. Briefly, CASP1 were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100uL were then transferred to IL1a-coated microtiter wells and incubated for 2h at 37 degree C. Wells were washed with PBST and incubated for 1h with anti-CASP1 pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37 degree C. Finally, add 50uL stop solution to the wells and read at 450nm immediately. The binding activity of of CASP1 and IL1a was shown in Figure 1, and this effect was in a dose dependent manner.)

Western Blot (WB) (Figure 1. Western blot analysis of Bag1 using anti-Bag1 antibody (MBS1750768).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: rat kidney tissue lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Bag1 antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for Bag1 at approximately 39KD. The expected band size for Bag1 is at 39KD.)
