Principle of the Assay: The MyBioSource' Rat EGF ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Rat EGF in Cell Culture Supernatants, Serum, Plasma, Tissue Homogenates, Urine. This assay employs an antibody specific for Rat EGF coated on a 96-well plate. Standards and samples are pipetted into the wells and EGF present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat EGF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of EGF bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Epidermal growth factor or EGF is a growth factor that plays an important role in the regulation of cell growth, proliferation, and differentiation by binding to its receptor EGFR. Human EGF is a 6045-Da protein with 53 amino acid residues and three intramolecular disulfide bonds. EGF acts by binding with high affinity to epidermal growth factor receptor (EGFR) on the cell surface and stimulating the intrinsic protein-tyrosine kinase activity of the receptor. EGF results in cellular proliferation, differentiation, and survival. It also has a profound effect on the differentiation of specific cells in vivo and is a potent mitogenic factor for a variety of cultured cells of both ectodermal and mesodermal origin. EGF has strong expression in kidney, salivary gland, cerebrum, and prostate, moderate expression in trachea and thyroid, and low expression in bone marrow, heart, spleen, thymus, uterus, and colon. No expression was detected in adrenal gland, liver, lung, cerebellum, placenta, and small intestine.

NCBI Description

binds the epidermal growth factor receptor; may play a role in MAP kinase mediated signaling pathways [RGD, Feb 2006]

Uniprot Description

EGF: EGF stimulates the growth of various epidermal and epithelial tissues in vivo and in vitro and of some fibroblasts in cell culture. Magnesiotropic hormone that stimulates magnesium reabsorption in the renal distal convoluted tubule via engagement of EGFR and activation of the magnesium channel TRPM6. Defects in EGF are the cause of hypomagnesemia type 4 (HOMG4); also known as renal hypomagnesemia normocalciuric. HOMG4 is a disorder characterized by massive renal hypomagnesemia and normal levels of serum calcium and calcium excretion. Clinical features include seizures, mild-to mederate psychomotor retardation, and brisk tendon reflexes. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Rat EGF egf (Catalog #AAA824918) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA824918 ELISA Kit recognizes Rat EGF. AAA Biotech's EGF can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (SE). Researchers should empirically determine the suitability of the EGF egf for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "EGF, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
