Isotype
IgG
Specificity
This item recognises an epitope within the N-terminal region (NT) of mouse TRIAD3A, a ubiquitously expressed E3 ubiquitin-protein ligase which plays an important role in the innate immune response.

Regulation of the level and duration of Toll-like receptor (TLR) activation is vital in controlling a pro-inflammatory response, to prevent harm to the host. Several proteins involved in the regulation of TLR signaling have been described including ST2 and IL-1R-associated kinase M, along with many other adaptor proteins of the MyD88 family, which mediate TLR signal transduction. TRIAD3A, identified as a RING finger member of the E3 ligase family of proteins, binds to the cytoplasmic tail of several TLRs, notably TLR4 and TLR9, resulting in ubiquitination and proteolytic degradation, thereby regulating their expression. TRIAD3A does not appear to regulate the expression of TLR2.

Uniprot Description

TRIAD3: Isoform 1 acts as an E3 ubiquitin ligase, which accepts ubiquitin from specific E2 ubiquitin-conjugating enzymes, and then transfers it to substrates promoting their degradation by the proteasome. Promotes degradation of TRAF3, TLR4 and TLR9. Contributes to the regulation of antiviral responses. Down- regulates activation of NF-kappa-B, IRF3 activation and IFNB production. Isoform 3/ZIN inhibits TNF and IL-1 mediated activation of NF-kappa-B. Promotes TNF and RIP mediated apoptosis. 3 isoforms of the human protein are produced by alternative splicing.

The TRIAD3A rnf216 (Catalog #AAA221309) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's TRIAD3A can be used in a range of immunoassay formats including, but not limited to, Immunohistology Paraffin*, Western Blot (WB). Western Blot: This item detects a band of approximately 95kDa in mouse heart cell lysates. Immunohistology - Paraffin: Application Note: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose. Western Blotting: Minimum Dilution: 0.5; Maximum Dilution: 1.0ug/ml. Researchers should empirically determine the suitability of the TRIAD3A rnf216 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TRIAD3A, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
