Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-NT-3 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-NT-3 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the NT-3 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of NT-3 can be calculated!!Background/Introduction: Neurotrophin-3 is a neurotrophic factor in the NGF (Nerve Growth Factor) family of neurotrophins. It is encoded by the NTF3 gene, which localized to 12p13. It is a protein growth factor which has activity on certain neurons of the peripheral and central nervous system; it helps to support the survival and differentiation of existing neurons, and encourages the growth and differentiation of new neurons and synapses. Ramer et al. (2000) concluded that neurotrophic factor treatment may serve as a viable treatment in promoting recovery from root avulsion injuries. Although the vast majority of neurons in the mammalian brain are formed prenatally, parts of the adult brain retain the ability to grow new neurons from neural stem cells; a process known as neurogenesis.

The protein encoded by this gene is a member of the neurotrophin family, that controls survival and differentiation of mammalian neurons. This protein is closely related to both nerve growth factor and brain-derived neurotrophic factor. It may be involved in the maintenance of the adult nervous system, and may affect development of neurons in the embryo when it is expressed in human placenta. NTF3-deficient mice generated by gene targeting display severe movement defects of the limbs.
