Application Notes
IHC-P dilution 1:100 - 1:200

IHC-Protocol - Instruction Manual:
1. Deparaffinize the section in 3 changes of xylene, 5 minutes each.
2. Wash the section in 96%, 80% and 70% ethyl alcohol for 10 minutes each.
3. Rinse in distilled water.
4. Block the endogenous peroxidase by incubating the tissue in 3% hydrogen peroxide (H2O2) for 10 minutes.
5. Wash in distilled water for 5 minutes.
6. Wash in 0.05 M Tris-HCl, pH 7.6 buffer supplemented with 1% of Tween-20 (buffer A) for 5 minutes.
7. CONCENTRATED:
Incubate the section with primary antibody at the dilution 1:100 - 1:200 for 1 hour in the closed wet chamber.
READY TO USE (RTU):
Incubate the section with primary antibody (ready to use) for 1 hour in a closed wet chamber.
8. Wash twice 5 minutes with buffer A.
9. Apply the secondary antibody (the protocol depends on the supplier), and proceed to standard immunohistochemistry protocol (HRP - Peroxide - DAB). Micropolymer-HRP detection kit rabbit/mouse dual is suggested.
10. Wash twice 5 minutes with buffer A.
11. Apply the chromogen (DAB), 1 - 3 minutes.
12. Wash in water for 10 minutes.
13. Stain in hematoxylin for 5 minutes.
14. Wash in water for 10 minutes.
15. Dehydrate the section in 2 changes of 96% ethyl alcohol for 5 minutes each.
16. Wash the section in 2 changes of xylene for 2 minutes each.
17. Mount the slide for observation.

Peptide derived from N-terminal sequence of human S100-A1 protein. Antibody recognizes the epitope between Val55 - Glu74.

Precautions
1. We strongly recommend to use Primary Antibody Diluent (please inquire), eventually alternative diluent (containing protease free BSA at the concentrations > 1mg/ml) for dilution of concentrated antibodies, otherwise the warranty might be voided.
2. Centrifuge the vial before use.
3. Intended for professional use in laboratories.
4. Do not use after expiration date stamped on vial label.
5. Avoid contamination of the reagent.
6. Any discrepancies in the recommended procedures stated in the working protocol may affect the final results.
7. The reagent contains sodium azide (NaN3) which is highly toxic in higher concentrations. The concentration in the reagent (0.05%) is not considered as hazardous.
8. Disposal of waste material must be conducted in accordance with local regulations.
9. Wear appropriate Personal Protective Equipment to avoid contact with eyes and skin.

NCBI Description

The protein encoded by this gene is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 genes include at least 13 members which are located as a cluster on chromosome 1q21. This protein may function in stimulation of Ca2+-induced Ca2+ release, inhibition of microtubule assembly, and inhibition of protein kinase C-mediated phosphorylation. Reduced expression of this protein has been implicated in cardiomyopathies. [provided by RefSeq, Jul 2008]

Function: Weakly binds calcium but binds zinc very tightly-distinct binding sites with different affinities exist for both ions on each monomer. Physiological concentrations of potassium ion antagonize the binding of both divalent cations, especially affecting high-affinity calcium-binding sites. May mediate calcium-dependent regulation on many physiological processes by interacting with other proteins, such as TPR-containing proteins, and modulating their activity. Ref.13

Subunit structure: Dimer of either two alpha chains, or two beta chains, or one alpha and one beta chain. Also forms heterodimers with S100P. Interacts with AGER and CAPZA1. Interacts with FKBP4. Interacts with RYR1 and RYR2. Interacts with CACYBP in a calcium-dependent manner. Interacts with PPP5C (via TPR repeats); the interaction is calcium-dependent and modulates PPP5C activity. Subcellular location: Cytoplasm.

Tissue specificity: Highly prevalent in heart. Also found in lesser quantities in skeletal muscle and brain.

Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "S-100, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening.
