Application Notes
10C2 Nestin is useful for immunocytochemistry, immunohistochemistry on frozen and paraffin-embedded tissues, and Western blot. Optimal antibody dilution should be determined by titRation; recommended range is 1:100 to 1:200 for immunocytochemistry and immunohistochemistry with a fluorescein or rhodamine conjugated anti-Mouse secondary antibody, and 1:1000 to 1:5000 for Western blot appliCations using chemoluminescent detection.
Source Note
Nestin (clone 10C2) is a monoclonal antibody derived by traditional hybridoma technology. A 150-amino-acid fragment from the cloned Human nestin was conjugated to glutathione S-transferase and used as an immunogen.
Preparation and Storage
Can be stored at -20 degree C to -70 degree C for long term storage. Once thawed, store at 4 degree C, do not refreeze.

Nestin is a Class IV intermediate filament protein abundantly produced in the developing central nervous system (CNS) and in early embryonic neuroepithelial stem cells. This cytoskeletal protein has been widely used as a predominant marker for stem/progenitor cells, glioma cells, and tumor endothelial cells. Nestin is an intermediate filament protein that was first identified with a monoclonal antibody by Hockfield and McKay (1985) [PubMed 4078630]. It is expressed predominantly in stem cells of the central nervous system in the neural tube. Upon terminal neural differentiation, nestin is downregulated and replaced by neurofilaments.[supplied by OMIM]
