Application Notes
Applications Validated:
WB: 1-2ug/ml dilution
For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all blocking solutions may also reduce background.
Immunogen
This Caspase-14 antibody was raised against a synthetic peptide corresponding to amino acid residues 2-18 of mouse Caspase-14.
Preparation and Storage
Some products may be shipped at room temperature. This will not affect their stability or performance. Avoid repeated freeze/thaw cycles by aliquoting items into single-use fractions for storage at -20 degree C for up to 2 years. Keep all reagents on ice when not in storage.1ug/ul

Related Product Information for anti-Casp14 antibody
Short Description: Our Caspase-14 antibody (mAb)(Clone 70A1426) was raised in a Mouse host. It has been validated for use in Western blot; it has been shown to react with Human and Mouse samples.

Background: Caspase-14 (CASP14 or apoptosis-related cysteine peptidase 14) is thought to be a non-apoptotic caspase involved in epidermal differentiation. Caspase-14 has been shown to be processed and activated by Caspase-8 and Caspase 10 in vitro, and also by TNF-Related Apoptosis-Inducing Ligand (TRAIL) in vivo.

Uniprot Description

CASP14: Believed to be a non-apoptotic caspase which is involved in epidermal differentiation. Seems to play a role in keratinocyte differentiation and cornification. Probably regulates maturation of the epidermis by proteolytically processing filaggrin. Complex of unprocessed caspase-14 and processed 19 kDa (p19) and 10 kDa (p10) subunits. In undifferentiated keratinocytes under postconfluency growth conditions (in vitro). Expressed in keratinocytes of adult skin suprabasal layers (from spinous layers to the stratum granulosum and stratum corneum). Expressed in keratinocytes of hair shaft and sebaceous glands. In psoriatic skin only expressed at very low levels. Belongs to the peptidase C14A family.

Protein type: EC 3.4.22.-; Protease

Cellular Component: keratin filament; cytoplasm; nucleus

Molecular Function: peptidase activity; hydrolase activity; cysteine-type peptidase activity

Biological Process: caspase activation; DNA damage response, signal transduction resulting in induction of apoptosis; apoptosis; cell differentiation; proteolysis

Product Notes

The Casp14 casp14 (Catalog #AAA388688) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Caspase-14 antibody (mAb)(Clone 70A1426) reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's Caspase-14 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Applications Validated: WB: 1-2ug/ml dilution For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all blocking solutions may also reduce background. Researchers should empirically determine the suitability of the Casp14 casp14 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Caspase-14, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
