LX-2343 is an non-ATP competitive PI3K inhibitor that negatively regulates AKT/mTOR signaling, thus promoting autophagy and increasing Abeta clearance, also inhibits BACE1 enzymatic activity with IC50 of 11.43uM; reduced Abeta production involving JNK/APPThr668 pathway inhibition, LX2343 (5-20uM) dose-dependently decreases Abeta accumulation in HEK293-APPsw and CHO-APP cells, and promotes Abeta clearance in SH-SY5Y cells and primary astrocytes; significantly ameliorates cognitive deficits and markedly ameliorates the Abeta pathology in brains in APP/PS1 transgenic mice.
