Principle of the Assay: The Picokine Human IL10 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human IL10 with a 96-well strip plate that is pre-coated with antibody specific for IL10. The detection antibody is a biotinylated antibody specific for IL10. The capture antibody is monoclonal antibody from mouse, the detection antibody is monoclonal antibody from mouse. The kit contains recombinant Human IL10 with immunogen: Expression system for standard: E.coli; Immunogen sequence: S19-N178. The kit is analytically validated with ready to use reagents. To measure Human IL10, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly propotional to Human IL10 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human IL10 in the sample!!Background/Introduction: Interleukin-10, also called cytokine synthesis inhibitory factor, is implicated in tumorigenesis, and it has been shown that polymorphisms in its gene promoter correlate with differential amounts of production. It suppresses the ability of CD4+ T cells, but not CD8+ T cells, to migrate in response to IL-8. NCBI Description

This gene encodes a cytokine receptor that specifically binds interleukin 15 (IL15) with high affinity. The receptors of IL15 and IL2 share two subunits, IL2R beta and IL2R gamma. This forms the basis of many overlapping biological activities of IL15 and IL2. The protein encoded by this gene is structurally related to IL2R alpha, an additional IL2-specific alpha subunit necessary for high affinity IL2 binding. Unlike IL2RA, IL15RA is capable of binding IL15 with high affinity independent of other subunits, which suggests distinct roles between IL15 and IL2. This receptor is reported to enhance cell proliferation and expression of apoptosis inhibitor BCL2L1/BCL2-XL and BCL2.
