Principle of the Assay: The Picokine Human IL18R1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human IL18R1 with a 96-well strip plate that is pre-coated with antibody specific for IL18R1. The detection antibody is a biotinylated antibody specific for IL18R1. The capture antibody is monoclonal antibody from mouse, the detection antibody is polyclonal antibody from goat. The kit contains recombinant Human IL18R1 with immunogen: Expression system for standard: NSO; Immunogen sequence: C22-R329. The kit is analytically validated with ready to use reagents. To measure Human IL18R1, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Human IL18R1 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human IL18R1 in the sample!!Background/Introduction: The interleukin-18 receptor 1(IL-18R1), also known as CDw218a(cluster of differentiation w218a) or IL18RA, is an interleukin receptor of the immunoglobulin superfamily. The protein encoded by this gene is a cytokine receptor that belongs to the interleukin 1 receptor family. This gene along with four other members of the interleukin 1 receptor family, including IL1R2, IL1R1, ILRL2(IL-1Rrp2), and IL1RL1(T1/ST2), form a gene cluster on chromosome 2q. This receptor specifically binds interleukin 18(IL18), and is essential for IL18 mediated signal transduction. IFN-alpha and IL12 are reported to induce the expression of this receptor in NK and T cells.

NCBI Description

The protein encoded by this gene is a cytokine that belongs to the IL10 cytokine subfamily. This cytokine is found to be preferentially expressed in monocytes. It can bind the IL20 receptor complex and lead to the activation of the signal transducer and activator of transcription 3 (STAT3). A similar cytokine in mouse is reported to up-regulate the expression of IL6 and TNF-alpha and induce apoptosis, which suggests a role of this cytokine in inflammatory responses.
