Principle of the Assay: The MBS Picokine™ Human EPHA1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Human EPHA1 with a 96-well strip plate that is pre-coated with antibody specific for EPHA1. The detection antibody is a biotinylated antibody specific for EPHA1. The capture antibody is A monoclonal antibody from mouse, the detection antibody is a biotinylated detection polyclonal antibody from goat. The kit contains recombinant Human EPHA1 with immunogen: Expression system for standard: NSO, Immunogen sequence: K26-E547. The kit is analytically validated with ready-to-use reagents. To measure Human EPHA1, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The density of the yellow product is linearly proportional to Human EPHA1 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human EPHA1 in the sample.

Background/Introduction: EPHA1, EPH receptor A1, is a protein that in humans is encoded by the EPHA1 gene. This gene belongs to the ephrin receptor subfamily of the protein-tyrosine kinase family. The EPHA1 gene is mapped to 7q34-q35. EPH and EPH-related receptors have been implicated in mediating developmental events, particularly in the nervous system. Receptors in the EPH subfamily typically have a single kinase domain and an extracellular region containing a Cys-rich domain and 2 fibronectin type III repeats. The ephrin receptors are divided into 2 groups based on the similarity of their extracellular domain sequences and their affinities for binding ephrin-A and ephrin-B ligands. This gene is expressed in some human cancer cell lines and has been implicated in carcinogenesis.
