Principle of the Assay: The Picokine™ Rat Cntn1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Rat Cntn1 with a 96-well strip plate that is pre-coated with antibody specific for Cntn1. The detection antibody is a biotinylated antibody specific for Cntn1. The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated polyclonal antibody from goat. The kit contains recombinant Rat Cntn1 with immunogen: Expression system for standard: NSO, Immunogen sequence: E21-S1001. The kit is analytically validated with ready to use reagents. To measure Rat Cntn1, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Rat Cntn1 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Rat Cntn1 in the sample.!!Background/Introduction: Contactin 1, also known as CNTN1, is a protein which in humans is encoded by the CNTN1 gene. It is mapped to 7q35. The protein encoded by this gene is a member of the immunoglobulin superfamily. It is a glycosylphosphatidylinositol (GPI)-anchored neuronal membrane protein that functions as a cell adhesion molecule. It may play a role in the formation of axon connections in the developing nervous system. Two alternatively spliced transcript variants encoding different isoforms have been described for this gene.

NCBI Description

neural adhesion molecule [RGD, Feb 2006]

Uniprot Description

CNTN1: Contactins mediate cell surface interactions during nervous system development. Involved in the formation of paranodal axo-glial junctions in myelinated peripheral nerves and in the signaling between axons and myelinating glial cells via its association with CNTNAP1. Participates in oligodendrocytes generation by acting as a ligand of NOTCH1. Its association with NOTCH1 promotes NOTCH1 activation through the released notch intracellular domain (NICD) and subsequent translocation to the nucleus. Interaction with TNR induces a repulsion of neurons and an inhibition of neurite outgrowth. Defects in CNTN1 are the cause of Compton-North congenital myopathy (CNCM). CNCM is a familial lethal form of congenital onset muscle weakness, inherited in an autosomal-recessive fashion and characterized by a secondary loss of beta2-syntrophin and alpha-dystrobrevin from the muscle sarcolemma, central nervous system involvement, and fetal akinesia. Belongs to the immunoglobulin superfamily. Contactin family. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Rat CNTN1 cntn1 (Catalog #AAA1752567) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA1752567 ELISA Kit recognizes Rat CNTN1. It is sometimes possible for the material contained within the vial of "Contactin-1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
