Principle of the Assay: The Picokine™ Human CLEC14A Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human CLEC14A with a 96-well strip plate that is pre-coated with antibody specific for CLEC14A. The detection antibody is a biotinylated antibody specific for CLEC14A. The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated polyclonal antibody from goat. The kit contains recombinant Human CLEC14A with immunogen: Expression system for standard: NSO, Immunogen sequence: E22-A490. The kit is analytically validated with ready to use reagents. To measure Human CLEC14A, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Human CLEC14A in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human CLEC14A in the sample.!!Background/Introduction: CLEC14A (C-type lectin domain family 14 member A; also EGFR5) is a 51 kDa (predicted) member of the C-type lectin domain family of proteins. This gene is mapped to 14q21.1. This gene encodes a member of the C-type lectin/C-type lectin-like domain (CTL/CTLD) superfamily. Members of this family share a common protein fold and have diverse functions, such as cell adhesion, cell-cell signalling, glycoprotein turnover, and roles in inflammation and immune response. This family member plays a role in cell-cell adhesion and angiogenesis. It functions in filopodia formation, cell migration and tube formation. Due to its presence at higher levels in tumor endothelium than in normal tissue endothelium, it is considered to be a candidate for tumor vascular targeting.
