• Materials Provided: 1. Capture Antibody 0.5 mg/mL of rabbit anti-mTHSD1 monoclonal antibody (in PBS, pH 7.4). Dilute to a working concentration of 1ug/mL in PBS before coating. ; 2. Detection Antibody 0.25 mg/mL rabbit anti-mTHSD1 polyclonal antibody conjugated to horseradish-peroxidase (HRP) (in PBS, 50 % HRP-protector, pH 7.4, store at 4 degree C). Dilute to working concentration of 0.25ug/mL in detection antibody dilution buffer before use.3. Standard Each vial contains 95 ng of recombinant mTHSD1. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20 degree C to -80 degree C in a manual defrost freezer. A seven-point standard curve using 2-fold serial dilutions in sample dilution buffer, and a high standard of 4ng/mL is recommended.

Avoid repeated freeze-thaw cycles.; Detection Antibody: Store at 4 degree C and protect it from prolonged exposure to light for up to 6 months from date of receipt. DO NOT FREEZE!; Standard: Store lyophilized standard at -20 degree C to -80 degree C for up to 6 months from date of receipt. Aliquot and store the reconstituted Standard at -80 degree C for up to 1 month. Avoid repeated freeze-thaw cycles.

Thrombospondin type-1 domain-containing protein 1, also known as transmembrane molecule with thrombospondin module, THSD1 and TMTSP, is a single-pass type I membrane protein that contains one TSP type-1 domain. THSD1 is a multi-domain, multi-functional glycoprotein synthesized by many cells. Matricellular THSD1 modulates cell adhesion and proliferation. It is involved in angiogenesis, inflammation, wound healing and cancer. In vitro, nanomolar concentrations of Thrombospondin-1 are required to alter endothelial and vascular smooth muscle cell adhesion, proliferation, motility, and survival. As a major platelet protein, for a long time it was postulated to control hemostasis via platelet aggregate stabilization. THSD1 is a potent angiogenesis inhibitor, and down-regulation of THSD1 has been suggested to alter tumor growth by modulating angiogenesis in a variety of tumor types.
