Materials Provided
1. Capture Antibody 0.5 mg/mL of mouse anti-GRN monoclonal antibody (in PBS, pH 7.4). Dilute to a working concentration of 2ug/mL in CBS before coating.
2. Detection Antibody 0.1 mg/mL mouse anti-GRN monoclonal antibody conjugated to horseradish-peroxidase (HRP) (in PBS, 50 % HRP-Protector, pH 7.4). Dilute to working concentration of 0.5ug/mL in detection antibody dilution buffer before use.
3. Standard Each vial contains 70 ng of recombinant GRN. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20 degree C to -80 degree C in a manual defrost freezer. A seven-point standard curve using 2-fold serial dilutions in sample dilution buffer, and a high standard of 4ng/mL is recommended.
Preparation and Storage
Capture Antibody: Aliquot and store at -20 degree C to -80 degree C for up to 6 months from date of receipt. Avoid repeated freeze-thaw cycles.
Detection Antibody: Store at 4 degree C and protect it from prolonged exposure to light for up to 6 months from date of receipt. DO NOT FREEZE!
Standard: Store lyophilized standard at -20 degree C to -80 degree C for up to 6 months from date of receipt. Aliquot and store the reconstituted Standard at -80 degree C for up to 1 month. Avoid repeated freeze-thaw cycles.

Granulins are a family of secreted, glycosylated peptides that are cleaved from a single precursor protein with 7.5 repeats of a highly conserved 12-cysteine granulin/epithelin motif. The precursor protein, progranulin, is also called proepithelin and PC cell-derived growth factor. Cleavage of the signal peptide produces mature granulin which can be further cleaved into a variety of active, 6 kDa peptides. These smaller cleavage products are named granulin A, granulin B, granulin C, etc. Epithelins 1 and 2 are synonymous with granulins A and B, respectively. Both the peptides and intact granulin protein regulate cell growth. However, different members of the granulin protein family may act as inhibitors, stimulators, or have dual actions on cell growth. Granulin family members are important in normal development, wound healing, and tumorigenesis. Granulins have possible cytokine-like activity. They may play a role in inflammation, wound repair, and tissue remodeling. Granulin-4 promotes proliferation of the epithelial cell line A431 in culture while granulin-3 acts as an antagonist to granulin-4, inhibiting the growth. Granulin expression inhibited Tat transactivation, and tethering experiments showed that this effect was due, at least in part, to a direct action on cyclin T1 in the absence of Tat.

This Granulin Matched ELISA Antibody Pair Set, Human is a solid phase sandwich ELISA for quantitative determination of Human Granulin. It contains Human Granulin capture antibody, Human Granulin detector antibody (HRP) and a highly purified HEK293-expressed recombinant Human Granulin protein.
