Application Notes
Dilution: Flow cytometry: 50ug/ml; 10ul labels 1-2.5x10e5 cells in a total reaction volume 200ul. Tested on 4T1 mouse mammary carcinoma cells.
Western Blot: 0.1-0.2ug/ml. The detection limit is ~5ng/lane under non-reducing and reducing conditions. In this format, shows <5% crossreactivity with recombinant human Integrin alpha5 and recombinant mouse Integrin alpha5, and <1% crossreactivity with recombinant mouse Integrin alphaV.
Immunocytochemistry: 10ug/ml detects Integrin alpha8 in 4T1 mouse mammary carcinoma cells. Cells were fixed with PBS, 4% paraformaldehyde for 20 minutes at RT and blocked with PBS, 10% normal donkey serum, 0.1% Triton X-100, 1% BSA for 45 minutes at RT. After blocking, cells were incubated with diluted I7661-36V1 for 3 hours at RT followed by a fluorochrome-conjugated streptavidin at RT for 1 hour. Between each step, cells were washed with PBS, 0.1% BSA.

Recombinant corresponding to aa38-1007 from the extracellular domain of mouse Integrin alpha8, expressed in CHO cells (NP_001001309).
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile TBS, 0.1% BSA. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Integrin alpha 8 is a 170-200kD member of the Integrin family of adhesion molecules. It forms an exclusive noncovalent heterodimer with Integrin b1. Alpha8beta1 promotes both cell adhesion and survival and is known to bind to fibronectin, the latency-associated peptide in latent TGFb1 and nephronectin. Mouse Integrin b8 is a 1025 aa type I transmembrane glycoprotein. It contains a 969aa extracellular region (aa38-1006) and a 29aa cytoplasmic domain. One isoform may exist that shows a truncation after Ala675 of the precursor. In the ECD, mouse Integrin a8 shares 96% and 90% aa sequence identity with rat and human Integrin a8 protein, respectively.

NCBI Description

Integrins are heterodimeric transmembrane receptor proteins that mediate numerous cellular processes including cell adhesion, cytoskeletal rearrangement, and activation of cell signaling pathways. Integrins are composed of alpha and beta subunits. This gene encodes the alpha 8 subunit of the heterodimeric integrin alpha8beta1 protein. The encoded protein is a single-pass type 1 membrane protein that contains multiple FG-GAP repeats. This repeat is predicted to fold into a beta propeller structure. This gene regulates the recruitment of mesenchymal cells into epithelial structures, mediates cell-cell interactions, and regulates neurite outgrowth of sensory and motor neurons. The integrin alpha8beta1 protein thus plays an important role in wound-healing and organogenesis. Mutations in this gene have been associated with renal hypodysplasia/aplasia-1 (RHDA1) and with several animal models of chronic kidney disease. Alternate splicing results in multiple transcript variants encoding distinct isoforms. [provided by RefSeq, Apr 2014]

It recognizes the sequence R-G-D in a wide array of ligands including TNC, FN1, SPP1 TGFB1, TGFB3 and VTN. NPNT is probably its functional ligand in kidney genesis. Neuronal receptor for TNC it mediates cell-cell interactions and regulates neurite outgrowth of sensory and motor neurons. Belongs to the integrin alpha chain family.
