Application Notes
Suitable for use in Flow Cytometry and Western Blot.
Dilution: Flow Cytometry: 50ug/ml; 10ul labeles 1-2.5x10e5 cells in a total reaction volume 200ul. Tested on bEnd.3 cells.
Western Blot: 0.1-0.2ug/ml; The detection limit is ~0.5ng/lane and 25ng/lane under non-reducing and reducing conditions, respectively. Shows less than 1% crossreactivity with recombinant human Thrombomodulin.

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile TBS, 0.1% BSA. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Thrombomodulin (TM; also fetomodulin and CD141) is a 110-120kD, endothelial cell-specific type I transmembrane glycoprotein. It binds thrombin, rendering it unavailable for clotting, and activates protein C, which also serves as an anticoagulant. Mouse Thrombomodulin/TM is 577 amino acids (aa) in length. It contains a 510aa extracellular domain (ECD) (aa17-517) that shows one C-type lectin domain (aa31-167) and six EGF-like regions (aa240-480). Thrombin binds to aa403-480. One isoform may exist that shows a deletion of the C-terminal 86 amino acids (aa492-577). Over aa17-517, mouse TM is 66%, 85% and 67% aa identical to human, rat and canine TM, respectively.

NCBI Description

The protein encoded by this intronless gene is an endothelial-specific type I membrane receptor that binds thrombin. This binding results in the activation of protein C, which degrades clotting factors Va and VIIIa and reduces the amount of thrombin generated. Mutations in this gene are a cause of thromboembolic disease, also known as inherited thrombophilia. [provided by RefSeq]

thrombomodulin: Thrombomodulin is a specific endothelial cell receptor that forms a 1:1 stoichiometric complex with thrombin. This complex is responsible for the conversion of protein C to the activated protein C (protein Ca). Once evolved, protein Ca scissions the activated cofactors of the coagulation mechanism, factor Va and factor VIIIa, and thereby reduces the amount of thrombin generated. Defects in THBD are the cause of thrombophilia due to thrombomodulin defect (THPH12). A hemostatic disorder characterized by a tendency to thrombosis. Defects in THBD are a cause of susceptibility to hemolytic uremic syndrome atypical type 6 (AHUS6). An atypical form of hemolytic uremic syndrome. It is a complex genetic disease characterized by microangiopathic hemolytic anemia, thrombocytopenia, renal failure and absence of episodes of enterocolitis and diarrhea. In contrast to typical hemolytic uremic syndrome, atypical forms have a poorer prognosis, with higher death rates and frequent progression to end-stage renal disease. Susceptibility to the development of atypical hemolytic uremic syndrome can be conferred by mutations in various components of or regulatory factors in the complement cascade system. Other genes may play a role in modifying the phenotype.
