Application Notes
Suitable for use in Direct ELISA, Western Blot and Flow Cytometry.
Dilution: Direct ELISA: 0.5-1.0ug/ml. The detection limit is ~0.2ng/well.
Western Blot: 0.1-0.2ug/ml. The detection limit is ~5ng/lane under non-reducing and reducing conditions.
Flow Cytometry: 25ug/ml. 10ul labels 1-2.5x10e5 cells in a total reaction volume <200ul. Tested using blood derived lymphocytes.

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Human Integrin Beta7 (also Integrin BetaP and M290 IEL Ag) is a 120kD member of the Integrin beta chain family of molecules. It is a type I transmembrane glycoprotein that contains a 704 amino acid (aa) extracellular domain (ECD) (aa20-723) and a 52aa cytoplasmic tail. The ECD contains one von Willebrand factor A domain (aa150-389) and four cysteine-rich EGF-like repeats. One splice variant exists that shows a deletion of aa501-648. Integrin Beta7 forms a nondisulfide linked heterodimer with alpha4 and alphaE integrins. Alpha4Beta7 binds fibronectin, VCAM-1 and MADCAM1, while alphaEBeta7 binds E-cadherin. The ECD of human Beta7 is 87% aa identical to the ECD of mouse Beta7.

NCBI Description

This gene encodes a protein that is a member of the integrin superfamily. Members of this family are adhesion receptors that function in signaling from the extracellular matrix to the cell. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. The encoded protein forms dimers with an alpha4 chain or an alphaE chain and plays a role in leukocyte adhesion. Dimerization with alpha4 forms a homing receptor for migration of lymphocytes to the intestinal mucosa and Peyer's patches. Dimerization with alphaE permits binding to the ligand epithelial cadherin, a calcium-dependent adhesion molecule. Alternate splicing results in multiple transcript variants. Additional alternatively spliced transcript variants of this gene have been described, but their full-length nature is not known. [provided by RefSeq, Sep 2013]

ITGB7: integrin alpha-4/beta-7 (Peyer's patches-specific homing receptor lpam-1) is expected to play a role in adhesive interactions of leukocytes. It is a receptor for fibronectin and recognizes one or more domains within the alternatively spliced CS-1 region of fibronectin. Integrin alpha-4/beta-7 is also a receptor for MADCAM1 and VCAM1. It recognizes the sequence L-D-T in MADCAM 1. Integrin alpha-e/beta-7 (HML-1) is a receptor for e-cadherin. Two alternatively spliced isoforms have been described.
