Application Notes
Suitable for use in Western Blot, Immunohistochemistry, ELISA and Flow Cytometry.
Dilution: Western Blot: 0.1-0.2ug/ml; The detection limit is 5ng/lane under non-reducing and 25ng/lane under reducing conditions.
Immunohistochemistry: 10ug/ml detects DLL1 in cryosections of E13.5 mouse embryos. Tissue was fixed overnight in 4% PFA+14% picric acid in PBS at 4 degree C. Sections were blocked with 10% donkey serum and 0.1% Triton in 1% BSA in PBS for 45 min at RT. After blocking, sections were incubated with D2727-20 for 3 hours at RT followed by fluorochrome-coupled anti-sheep lgG at RT for 1 hour. Between each step, sections were washed with 0.1% BSA in PBS.
Direct ELISA: 0.5-1.0ug/ml; The detection limit is 0.3ng/well.
Flow Cytometry: 25ug/ml; 10ul labels 1-2.5x10e5 cells in total reaction volume < 200ul. Validated using mouse splenocytes.
Optimal dilutions to be determined by researcher.
Immunogen
Recombinant protein corresponding to aa18-537 of rat Delta-like 1 expressed in NSO cells (Accession #: P97677). Species Sequence Homology: mouse (97%), human (91%).
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Delta-1 (also known as Delta-like 1, DLL1) is an 80-100kD vertebrate homolog of the Drosophila delta protein. Rat Delta-1 is a type I transmembrane protein that is 697aa in length. It contains a 520aa extracellular domain (ECD) and a 154aa cytoplasmic region. The ECD has one DSL segment (aa158-220) and eight consecutive EGF-like domains (aa225-515). Proteolytic processing of Delta-1 by ADAMs generates soluble intracellular and extracellular domains. Delta-1 binds to Notch and limits monocyte development while promoting T cell and DC development.

NCBI Description

This gene encodes a member of the delta protein ligand family. This family functions as Notch ligands that are characterized by a DSL domain, EGF repeats, and a transmembrane domain. Mutations in this gene cause autosomal recessive spondylocostal dysostosis 1. Two transcript variants encoding distinct isoforms have been identified for this gene. [provided by RefSeq, Jul 2008]
