Specificity
The assay recognizes natural and recombinant murine TNFalpha and rat TNFalpha. To define specificity of this ELISA, several proteins were tested for cross reactivity. There was no cross reactivity observed for the proteins tested human TNFalpha, murine IL-2, murine IL-6, murine IL-10, murine IL-23, murine IFNgamma, murine GM-CSF, murine CD138.

Intra-assay Precision
Reproducibility within the assay was evaluated in three independent experiments. Each assay was carried out with 6 replicates (3 duplicates) of 6 different samples containing different concentrations of TNFalpha. Data below show the mean concentration and the coefficient of variation for each sample. The overall intraassay coefficient of variation has been calculated to be 5%
Inter-assay Precision
Assay to assay reproducibility within one laboratory will be evaluated in three independent experiments. Each assay will be carried out with 6 replicates (3 duplicates) in 2 murine pooled serum, 2 in RPMI and 2 in standard diluent with samples containing different concentrations of TNFalpha. 2 standard curves were run on each plate. The calculated overall coefficient of variation was 9.4%.

Intended Uses: The Murine TNF-alpha ELISA is to be used for the in-vitro quantitative determination of murine tumor necrosis factor-alpha (mTNFalpha) in murine serum, buffered solutions or cell culture medium. The assay will recognize both natural and recombinant murine TNF-alpha.

Principle of the Assay: The murine TNF-alpha Kit is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (ELISA). A monoclonal antibody specific for mTNFalpha has been coated onto the wells of the microtiter strips provided. The antigen and a biotinylated polyclonal antibody specific for mTNFalpha are simultaneously incubated. Revelation step includes Streptavidin-Horse Radish peroxidase and TMB as chromogen.

NCBI Description

This protein belongs to the lipocalin family and is the specific carrier for retinol (vitamin A alcohol) in the blood. It delivers retinol from the liver stores to the peripheral tissues. In plasma, the RBP-retinol complex interacts with transthyretin which prevents its loss by filtration through the kidney glomeruli. A deficiency of vitamin A blocks secretion of the binding protein posttranslationally and results in defective delivery and supply to the epidermal cells. [provided by RefSeq, Jul 2008]

Defects in RBP4 are a cause of retinol-binding protein deficiency (RBP deficiency). This condition causes night vision problems. It produces a typical 'fundus xerophthalmicus', featuring a progressed atrophy of the retinal pigment epithelium. Belongs to the calycin superfamily. Lipocalin family.
