ELISA (EIA), Functional Assays (FN), Western Blot (WB)
Application Notes
ELISA: Recombinant human CD120b may be used as a standard for ELISA applications with either a purified human CD120b antibody or a biotinylated human CD120b antibody
Western Blot: Recombinant human CD120b may be used as the positive control in Western Blotting applications with either a purified human CD120b antibody or a biotinylated human CD120b antibody.
ELISA: Minimum Dilution: 0.2; Maximum Dilution: 0.4 ng/well
Functional Assays: Maximum Dilution: 0.125ug/ml
Western Blotting: Minimum Dilution: 1.5; Maximum Dilution: 3.0 ng/lane
Reconstitution
Reconstitute with 0.2ml distilled water. For extended storage the addition of 0.1% BSA is recommended. Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution.
Preparation
Buffer Solution
Phosphate buffered saline.
Activity
Determined by its inhibitory effect of the TNF-alpha mediated cytotoxicity in murine L-929 cells. The ED50 for this effect in the presence of 0.25 ng/ml of recombinant human TNF-alpha is 0.125 ug/ml.
Target Species
Human
Preparation and Storage
Prior to reconstitution store at 4 degree C. After reconstitution store at -20 degree C. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 3 months from date of reconstitution.
Related Product Information for CD120b active protein
Recombinant human CD120b is an 18.9kDa peptide comprised of 174 amino acids representing the soluble form of CD120b. Soluble human CD120b, otherwise known as sTNF receptor type II (sTNFRII), is a member of the tumour necrosis factor receptor (TNFR) superfamily, produced by proteolytic cleavage of the membrane form by the metalloprotease TACE. CD120b plays a key role in regulating the inflammatory response, acting as a receptor for both tumour necrosis factor alpha (TNF-alpha) and TNF-beta, and is also involved in programmed cell death and anti-viral activity. sTNFRII acts as an inhibitor of TNF-alpha induced responses, by acting as a decoy receptor.

NCBI Description

The protein encoded by this gene is a member of the TNF-receptor superfamily. This protein and TNF-receptor 1 form a heterocomplex that mediates the recruitment of two anti-apoptotic proteins, c-IAP1 and c-IAP2, which possess E3 ubiquitin ligase activity. The function of IAPs in TNF-receptor signalling is unknown, however, c-IAP1 is thought to potentiate TNF-induced apoptosis by the ubiquitination and degradation of TNF-receptor-associated factor 2, which mediates anti-apoptotic signals. Knockout studies in mice also suggest a role of this protein in protecting neurons from apoptosis by stimulating antioxidative pathways. [provided by RefSeq, Jul 2008]

Uniprot Description

TNF-R2: Receptor with high affinity for TNFSF2/TNF-alpha and approximately 5-fold lower affinity for homotrimeric TNFSF1/lymphotoxin-alpha. The TRAF1/TRAF2 complex recruits the apoptotic suppressors BIRC2 and BIRC3 to TNFRSF1B/TNFR2. This receptor mediates most of the metabolic effects of TNF-alpha. Isoform 2 blocks TNF-alpha-induced apoptosis, which suggests that it regulates TNF-alpha function by antagonizing its biological activity. Binds to TRAF2. Interacts with BMX. 2 isoforms of the human protein are produced by alternative splicing.
